链霉菌Streptomyces rosealbus次级代谢产物的分离和鉴定

Isolation and identification of secondary metabolites from Streptomyces rosealbus

摘要 目的研究三七根际土壤链霉菌Streptomyces rosealbus的次级代谢产物,以了解其次级代谢物结构多样性,为生物活性筛选提供单体化合物。方法利用MCI柱色谱、凝胶柱色谱、ODS柱色谱、半制备高效液相色谱、制备薄层色谱等手段进行分离,利用理化性质及波谱学数据并结合相关文献对所得化合物结构进行鉴定。结果从Streptomyces rosealbus发酵液的乙酸乙酯萃取物中分离得到6个化合物,分别鉴定为angucylone (1)、gephyromycin (2)、dehydrorabelomycin (3)、dehydroxyaquayamycin (4)、galtamycinone (5)、quanolironⅠ(6)。结论化合物1-6均为芳香聚酮类化合物,且首次从链霉菌Streptomyces rosealbus的次级代谢产物中分离得到,化合物1为新化合物。 Objective To study the secondary metabolites of Streptomyces rosealbus derived from the rhizosphere soil of Panax notoginseng, to understand the structural diversity of its secondary metabolites, and to provide monomer compounds for further biological activity screening.Methods The compounds were isolated by using MCI column chromatography, gel column chromatography, ODS column chromatography, semi-preparative HPLC and preparative TLC.The structures of the compounds were identified according to physical and chemical properties, spectroscopy methods combined with relevant literatures.Results Six compounds were isolated from the ethyl acetate partition of fermentation broth and were identified as anguylone(1),gephyromycin(2),dehydrorabelomycin(3),dehydroxyaquayamycin(4),galtamycinone(5),and quanoliron I(6) separately.Conclusion Compounds 1-6 are aromatic polyketones.It is the first time to obtain many kinds of aromatic polyketones from Streptomyces rosealbus,and compound 1 is identified as a new compound.
