摘要
目的:利用UPLC指纹图谱结合化学模式识别技术对昆布进行质量评价。方法:采用Waters Acquity UPLC BEH C18(100 mm×2.1 mm,1.7μm)色谱柱;以乙腈-水为流动相,梯度洗脱;流速为0.3 mL/min;检测波长为217 nm;柱温为30℃;进样量为4μL;建立昆布UPLC指纹图谱,并进行相似度评价、聚类分析及主成分分析。结果:建立的昆布UPLC指纹图谱确定33个共有峰,除了S4号样品外,其余样品相似度均大于0.850。聚类分析将11批样品聚为3类,S1号样品聚为一类,S4号样品聚为一类,S2、S3、S5~S11号样品聚为一类。主成分分析得到3个主成分因子的累计方差贡献率为83.653%,S4、S1号样品的主成分因子综合得分位于第1、2位,整体质量较好。结论:该研究建立的方法稳定、可靠、重复性好,可以更系统、全面地评价昆布的质量。
Objective:To evaluate the quality of thallus of Laminaria japonica by UPLC fingerprint and chemical pattern recognition technology.Methods:A Waters Acquity UPLC BEH C18(100 mm×2.1 mm,1.7μm)chromatographic column was used,the acetonitrile-water was used as mobile phase with gradient elution,the flow rate was 0.3 mL/min,the detection wavelength was 217 nm,the column temperature was 30℃,the injection volume was 4μL.The UPLC fingerprint of thallus of Laminaria japonica was established and the similarity evaluation,clustering analysis and principal component analysis were carried out.Results:The established thallus of Laminaria japonica UPLC fingerprint identified 33 common peaks,the similarity of all samples except S4 was greater than 0.850.In cluster analysis,11 batches of samples were clustered into three groups,S1 into one group,S4 into one group,S2,S3,S5 to S11 into one group.According to principal component analysis,the cumulative variance contribution rate of the three principal component factors was 83.653%,the comprehensive score of the principal component factors of samples S4 and S1 was in the first and second place,respectively,showing good overall quality.Conclusion:The method established in this study is stable,reliable and repeatable,which can evaluate the quality of thallus of Laminaria japonica more systematically and comprehensively.
