GC-FID内标法测定植物提取物中脂肪酸含量的方法研究 被引量:7

摘要 采用气相色谱仪建立了操作简单、检测用时短、成本低的脂肪酸含量检测方法,经验证该方法的精密度均在5%以内,方法稳定性较好,准确性高,满足了植物提取物中脂肪酸含量的检测需求。本方法选择十一碳酸甲酯为内标,根据GB 5009.257中校准因子计算方法建立以十一碳酸甲酯为内标的定量计算方法。该方法测定两种植物油中的脂肪酸含量,RSD在3%-7%范围内,加标回收率在93%-123%范围内。对植物提取物中脂肪酸含量进行分析,不同来源的样品脂肪酸组成具有特征性,其中:金阳青花椒中棕榈一烯酸含量高于武都红花椒及金阳红花椒,分别为11.78%、0.23%、1.79%;番茄皮提取物中棕榈酸含量高于番茄酱提取物,分别为11.29%和5.93%;印度TEJA辣椒油酸含量高于新疆甜椒,分别为13.28%和7.76%。基于该研究结论,初步说明脂肪酸组成有可能作为植物提取物不同来源鉴别的一项特征指标,需要其他学者持续开展研究。 In this paper,gas chromatograph was used to establish a fatty acid detection method with the advantages of simple operation,short detection time and low cost.It has been verified that the precision of the method is within 5%,and the method has good stability and high accuracy,which meets the requirements of plant extraction.The methyl undecanoate was selected as the internal standard,and calculation formula was established with methyl undecanoate as the internal standard according to the calculation method of calibration factor in GB 5009.257.The content of fatty acids in two kinds of vegetable oils was determined by this method.The RSD was in the range of 3%-7%,and the standard recovery was in the range of 93%-123%.The fatty acid content in plant extracts was analyzed.The fatty acid composition of samples from different sources possesses characteristic.Among them,the content of palm enoic acid in Jinyang Green Pepper is higher than Wudu Red Pepper and Jinyang Red Pepper,11.78%,0.23%and 1.79%respectively.The palmitic acid content in tomato skin extract is higher than that in tomato sauce extract,which is 11.29%and 5.93%respectively.The oleic acid content of Indian TEJA chili peppers is higher than Xinjiang sweet peppers,which is 13.28%and 7.76%respectively.Based on the conclusions of the study,it is preliminarily shown that the fatty acid composition may be used as a characteristic indicator for the identification of different sources of plant extracts,and other scholars are required to continue research.
