柴黄清胰活血方对重症急性胰腺炎模型大鼠肝脏氧化应激的影响 被引量:5

Effects of Chaihuang Qingyi Huoxue decoction on oxidative stress in the liver in the rat model of severe acute pancreatitis

摘要 目的探讨柴黄清胰活血方对重症急性胰腺炎(severe acute pancreatitis,SAP)模型大鼠肝脏氧化应激的影响。方法随机将128只SD大鼠分为空白组(Blank组)、假手术组(Sham组)、重症急性胰腺炎模型组(SAP组)和柴黄清胰活血方组(Chaihuang组)。经十二指肠乳头逆行胰胆管注射5%牛磺胆酸钠溶液制备SAP大鼠模型,麻醉苏醒后各组每隔6 h灌胃相应药物(即柴黄清胰活血方流浸膏药液)或生理盐水;术后6、12、24、48 h采集大鼠动脉血和肝组织,行HE染色观察肝脏病理并评分,生化法检测大鼠血清AMY、ALT、AST及肝组织SOD、CAT、GSH-Px、MDA的含量,Western Blot和RT-PCR法测定肝组织HO-1蛋白和mRNA的表达情况。结果与Sham组相比,Chaihuang和SAP组肝组织有明显病理改变;血清AMY、ALT、AST含量明显升高(P<0.05);肝组织SOD、CAT、GSH-Px水平降低(P<0.05),MDA活性升高(P<0.05);肝组织HO-1蛋白和mRNA表达水平升高(P<0.05)。与SAP组相比,Chaihuang组肝组织病理损伤明显减轻;血清AMY、ALT、AST含量降低(P<0.05);肝组织SOD、CAT、GSH-Px活性升高(P<0.05),MDA水平降低(P<0.05),HO-1蛋白和mRNA表达水平升高(P<0.05)。结论柴黄清胰活血方对SAP模型大鼠肝损伤具有保护作用,其作用机制可能与上调HO-1的表达,抑制氧化应激反应有关。 Objective To explore the effects of Chaihuang Qingyi Huoxue decoction on oxidative stress in the liver in the rat model of severe acute pancreatitis(SAP).Methods A total of 128 Sprague-Dawley rats were randomly divided into four groups:Blank group,Sham group,SAP group,and Chaihuang group.A rat model of SAP was established by retrograde injection of 5%sodium taurocholate solution into the pancreaticobiliary duct through the duodenal papilla.After awakening from anesthesia,the rats in each group were given corresponding Chaihuang Qingyi Huoxue decoction or normal saline every 6 hours.The arterial blood and hepatic tissue of the rats were collected at 6,12,24,and 48 h after operation.HE staining was performed to assess the pathological changes of hepatic tissue.Biochemical assays were conducted to measure the levels of serum amylase(AMY),alanine aminotransferase(ALT),and aspartate aminotransferase(AST)as well as the content of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px),and malondialdehyde(MDA)in hepatic tissue.RT-PCR was used to determine the mRNA expression of heme oxygenase-1(HO-1),and Western blot was used to measure the protein expression of HO-1.Results Compared with the Sham group,the Chaihuang group and SAP group showed significant pathological changes,significantly increased levels of serum AMY,ALT,and AST(P<0.05),significantly decreased levels of SOD,CAT,and GSH-Px in the liver tissue(P<0.05),significantly increased MDA activity(P<0.05),and significantly increased expression levels of HO-1 protein and mRNA in hepatic tissue(P<0.05).Compared with the SAP group,the Chaihuang group showed significantly reduced pathological injuries of the liver tissue,significantly decreased levels of serum AMY,ALT,and AST(P<0.05),significantly increased activity of SOD,CAT,and GSH-Px in the liver tissue(P<0.05),significantly reduced MDA activity(P<0.05),and significantly increased expression levels of HO-1 protein and mRNA(P<0.05).Conclusion Chaihuang Qingyi Huoxue decoction has a protective effect against liver injury in a SAP model in rats,and the mechanism may be related to up-regulation of HO-1 expression and inhibition of oxidative stress.
