摘要
为了获得产后卵巢静止奶牛血浆差异代谢物并明确其在卵巢静止的发生中的作用,本试验选取产后45~60d的卵巢静止组奶牛和发情组奶牛各15头,应用GC-TOF-MS技术分析两组奶牛血浆代谢谱,结合多元统计分析和单变量分析,筛选两组奶牛血浆差异代谢物,再经代谢通路分析,探究差异代谢物与奶牛卵巢静止的关系及其作用。结果显示:两组奶牛血浆GC-TOF-MS分析共筛选出16种血浆差异代谢物。其中与发情组相比,卵巢静止组奶牛有9种差异代谢物明显升高,7种明显下降。经代谢通路分析发现,所获得的奶牛卵巢静止血浆差异代谢物主要参与乙醛酸和二羧酸代谢,β-丙氨酸代谢,甘油脂代谢,果糖和甘露糖代谢,丙酸代谢,谷胱甘肽代谢,甘氨酸、丝氨酸和苏氨酸代谢,类固醇生物合成和嘧啶代谢等。结果表明,奶牛产后糖代谢、脂类代谢、氨基酸代谢和谷胱甘肽代谢等异常会阻碍卵泡的发育,从而导致奶牛产后发生卵巢静止。
The purpose of this study is to obtain the plasma differential metabolites of dairy cows with inactive ovaries and to clarify their role in the occurrence of inactive ovaries.Fifteen inactive ovaries cows and 15estrus cows between days 45and 60postpartum were selected.GC-TOF-MS techniques were used to obtain the plasma metabolic profiles of the two groups of cows,and the plasma differential metabolites of the two groups of cows were screened by multivariate statistical analysis and univariate analysis,then the relationship and functions of differential metabolites of cows with inactive ovaries were explored through metabolic pathway analysis.The results showed that sixteen plasma differential metabolites were screened from two groups of cows by GC-TOFMS analysis.Among them,nine differential metabolites were significantly increased in the cows with inactive ovaries compared with the estrus cows,seven differential metabolites were significantly decreased in the cows with inactive ovaries.The metabolic pathway analysis found that the plasma differential metabolites of dairy cow with inactive ovaries obtained by GC-TOF-MS techniques were involved in glyoxylate and dicarboxylate metabolism,β-alanine metabolism,glycerolipid metabolism,fructose and mannose metabolism,propionic acid metabolism,glutathione metabolism,glycine,serine and threonine metabolism,steroid biosynthesis metabolism and pyrimidine metabolism.The results indicated that the abnormalities in postpartum glycose metabolism,lipid metabolism,amino acid metabolism,and glutathione metabolism in dairy cows will impede the development of follicles,which can lead to postpartum inactive ovaries in dairy cows.
