绵羊副结核分枝杆菌的分离与分子分型 被引量:8

摘要 为了解绵羊副结核病的病原学特征,本研究对PCR检测阳性的绵羊肠系膜淋巴结样品开展细菌分离培养,对分离株进行形态观察,结果显示肠系膜淋巴结样品经培养后出现乳白色菌落;对分离菌株进行抗酸染色镜检后确定为抗酸染色阳性杆菌;PCR扩增该菌IS900基因与亚型分型片段,结果显示,IS900基因及亚型分型检测为Ⅱ型(牛型)副结核分枝杆菌(MAP);多位点短重复序列(MLSSR)和分枝杆菌散在分布重复单位-可变数目串联重复序列(MIRU-VNTR)分子分型结果显示,MLSSR和MIRU-VNTR分子分型数字代码分别为7+555555555和32632129。以上结果表明本研究分离的MAP是一种未曾报道的新基因型菌株,为首次在我国分离的绵羊源II型MAP。本研究为进一步开展绵羊MAP病原学和流行病学调查提供了参考资料。 To understand the etiological characteristics of sheep Mycobacterium avium subspecies paratuberculosis(MAP),the sheep mesenteric lymph node sample positive for PCR detection was carried out bacteria isolation,the isolate was detected by the colony morphology observation,and the results showed that milky white colonies appeared after the mesenteric lymph node sample was cultured.The acid-fast staining and microscopic examination showed that the isolate was a positive bacillus,PCR amplification of the IS900 gene and subtype fragment showed that the IS900 and subtype genes were detected as type Ⅱ(cattle type)MAP.Multilocus short sequence repeat(MLSSR)and mycobacterial interspersed repetitive unit-variable number of tandem repeat(MIRUVNTR)molecular typing showed that the numerical codes for MLSSR and MIRU-VNTR molecular typing were 7+555555555 and 32632129,respectively.The above results showed that the isolate was a novel genotype that had never been reported,indicating isolation of a type II(cattle type)MAP from sheep for the first time in China.The study provides reference for further etiological and epidemiological investigation of sheep MAP.
