摘要 目的确定红细胞保养液(MAP)混悬洗涤红细胞的容量范围及最佳保存期限。方法选取2019年6月至2020年3月该中心制备的MAP混悬洗涤红细胞107袋,称取血液重量后将其悬挂于4℃冰箱中保存,分别储存7、14、21、35 d后观察其外观并取样,检测其溶血率。于储存35 d测定上清蛋白、血红蛋白含量及比重等。结果107袋MAP混悬洗涤红细胞比重为1.048~1.063,平均1.058±0.003;1 U洗涤红细胞容量为133~160 mL,2 U洗涤红细胞容量为260~312 mL。21 d溶血率明显高于14 d,35 d溶血率明显高于21 d,差异均有统计学意义(P<0.05)。21 d血液外观70%出现轻度溶血。其他各项指标均符合国家质量标准。结论MAP混悬洗涤红细胞35 d后质量控制符合《全血及成分血质量要求(GB18469-2012)》,但随着储存时间的延长溶血率逐渐升高,上清液颜色逐渐加深,故将洗涤红细胞保存期确定为14 d。 Objective To determine the volume range and best shelf life of MAP suspension washed red blood cells.Methods One hundred and seven bags of MAP suspension washed red blood cells prepared by this center from June 2019 to March 2020 were selected,weighed and suspended in the refrigerator at 4℃for preservation.After the storage for 7,14,21,35 d,the appearance was observed and the sample was taken.The hemolysis rate was detected.The content of supernatant protein,hemoglobin and specific gravity were measured on 35 d of storage.Results The specific gravity of MAP suspension washed red blood cells in 107 bags was 1.048-1.063,with an averaged of 1.058±0.003.The volume of 1 U washed red blood cells was 133-160 mL,which of 2 U washed red blood cells was 260-312 mL.The hemolysis rate on 21 d was significantly higher than that on 14 d,the hemolysis rate on 35 d was significantly higher than that on 21 d,and the differences were statistically significant(P<0.05).70%of the blood appearance on 21 d showed mild hemolysis.The other indicators all met the national quality standard.Conclusion The quality control of MAP suspension washed red blood cells after 35 d meets the Quality Requirement of Whole Blood and Blood Components(GB19469-2012),but with the storage time increase,the hemolysis rate is gradually increased,the supernatant color is gradually deepened,therefore the retention period of washed red blood cells is determined as 14 d.
