摘要
目的:比较改良的体内注射秋水仙素制备法和空气干燥直接制备法制作小鼠生精细胞减数分裂中期染色体标本的优劣。方法:选用8~12周龄雄性Balb/c小鼠,分别采用改良的体内注射秋水仙素制备法和空气干燥直接制备法进行减数分裂中期染色体制备,并用Giemsa染色。光镜下观察小鼠生精细胞减数分裂不同阶段分裂相细胞染色体形态并计数,计算各分裂相细胞的检出率。结果:体内注射秋水仙素制备法和空气干燥直接制备法对生精细胞减数分裂中期染色体核型的检出率分别为10.37%和6.46%。秋水仙素处理小鼠的最适浓度是4 mg/kg,体内注射秋水仙素制备法制得的核型分裂相比空气干燥直接制备法多,直接制备法得到的染色体核型边缘清晰,更便于计数。结论:体内注射秋水仙素制备法的优势在于核型分裂相多,而空气干燥直接制备法的优势在于染色体核型边缘清晰,更便于计数,可根据不同实验需求进行选择。
Objective:To compare the advantages and disadvantages of the modified colchicine injection and the direct air drying methods for preparing meiotic metaphase chromosome of mouse spermatogenic cells.Methods:Male Balb/c mice aged 8 to 12 weeks were used to prepare meiotic metaphase chromosomes by the modified colchicine injection and air drying,respectively,and stained with Giemsa.Under light microscope,the chromosome morphology was observed and the mitotic phase cells were counted at different stages of meiosis in spermatogenic cells,and the rate of cells in each phase was calculated.Results:The detection rate of chromosome karyotypes in meiotic metaphase of spermatogenic cells by two methods was 10.37%and 6.46%respectively.The optimum concentration of colchicine for the mice was 4 mg/kg,and the karyotype obtained by injection of colchicine in vivo was more than that by direct preparation.The chromosome karyotype obtained by direct preparation had clear edges and was easier to count.Conclusion:Colchicine injection in vivo can obtain more karyotypes,while air drying direct preparation can obtain clear chromosome karyotype edge,which is easier to count,and can be selected according to the different experimental requirements.
