摘要
目的:使冰菖散吸嗅剂更加科学规范并验证其对轻度认知障碍(Mild cognitive impairment,MCI)模型小鼠的干预效果。方法:采用正交试验法,以挥发油的提取量为考察指标,优选挥发油最佳提取工艺条件,并采用GC-MS技术分析其主要化学成分,再加入冰片制备成冰菖散吸嗅剂,经鼻给药两周后检测其对MCI小鼠的筑巢行为以及β-淀粉样蛋白(Amyloidβ-protein,Aβ)、肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)等指标的影响。结果:(1)最佳提取工艺为:药材加入17.5倍水量,2%NaCl,50℃水浴3 h;经GC-MS分析,冰菖散混合挥发油成分中丁香酚的相对含量超过一半。(2)冰菖散吸嗅剂显著提高5.5月龄MCI小鼠的筑巢质量。(3)中药组(冰菖散嗅吸剂)血清TNF-α浓度显著降低,与模型组相比差异非常显著(P<0.001)。(4)病理切片显示:5.5月龄时,模型组MCI模型小鼠的脑皮层和海马均出现Aβ斑块,中药组(冰菖散嗅吸剂)皮层以及海马均没有Aβ沉积,而西药组(多奈哌齐)只有皮层中存在少量Aβ沉积,与模型组相比差异非常显著(P<0.001)。结论:优选出的提取工艺出油量较高,且稳定可行;冰菖散吸嗅剂经鼻给药可以减少Aβ沉积、降低血清TNF-α浓度从而提高5.5月龄MCI模型小鼠的认知功能。
Objective:To make the sniff-form Bingchangsan more scientific and normative and verify its intervention effects on MCI model mice.Methods:Taking the extraction amount of volatile oil as the index,optimized the extraction process conditions of volatile oil by orthogonal test,and analyzed its main chemical constituents by GC-MS technology.The sniff-form Bingchangsan was prepared by adding borneol,and after two weeks of intranasal administration,its effects were detected that on the nesting behavior of MCI model mice,and the Aβ,TNF-α,etc.Results:(1)The optimized extraction conditions were:the water was 17.5 times,2%NaCl,50℃water bath 3 h;The relative content of Eugenol in the volatile oil of Bingchangsan was more than half.(2)The sniff-form Bingchangsan significantly improved the nesting quality of 5.5-month-old MCI model mice.(3)The serum TNF-αconcentration in the sniff-form Bingchangsan group decreased very significantly,compared with the model group(P<0.001).(4)The pathological sections showed that:At the age of 5.5 months,in the model group,there were Aβplaques both in the cortex and hippocampus of the MCI model mice.There were no Aβdeposition both in the cortex and hippocampus of the sniff-form Bingchangsan group,while in the Donepezil group there was only a small amount of Aβdeposition in the cortex,which was highly statistically significant compared with the model group(P<0.001).Conclusion:The optimum extraction process has higher oil yield,and is stable and feasible.The sniff-form Bingchangsan can improve cognitive function of 5.5-month-old MCI model mice by reducing Aβdeposition and the serum TNF-αconcentration.
