摘要
随着黄粉虫养殖规模的扩大,废弃物中含有大量的黄粉虫虫蜕,而虫蜕中含有不易降解的几丁质。为缓解环境污染,实现黄粉虫养殖废弃物中虫蜕的资源化利用,以自然填埋黄粉虫虫蜕1 a的土壤为材料,采用透明圈法筛选几丁质降解菌(命名为Wn),经16S rDNA同源进化关系鉴定种属,最后通过单因素试验下的酶活性确定最优培养条件。结果表明,Wn为类芽孢杆菌属菌株,在以8 g/L的胶体几丁质为唯一碳源、装液量为1/3(50 mL/150 mL)、pH值为7.5、温度为30℃的发酵培养液中,培养96 h时的酶活性最大,为0.155 U/mL。研究结果可为黄粉虫废物利用和生物防治奠定基础。
With the expansion of the breeding scale of Tenebrio molitor L.,there are a lot of Tenebrio molitor L.molts in the waste,and the molting contains chitin which is not easy to degrade.To alleviate the environmental pollution and realize the resource utilization of molting in Tenebrio molitor L.breeding waste,the chitin-degrading bacterium named Wn were isolated from the soil of natural landfill of Tenebrio molitor L.molts for one year by transparent circle method and identified by 16S rDNA homologous evolutionary relationship.The optimal culture conditions were determined by enzyme activity under single factor test.The results showed that Wn was Paenibacillus sp.strain.In the fermentation medium with 8 g/L colloidal chitin as the sole carbon source,50 mL of liquid volume,pH 7.5,30℃,the enzyme activity was the highest(0.155 U/mL)at 96 h of culture.The results laid a foundation for the utilization of Tenebrio molitor L.waste and biological control.
