摘要
目的建立高效液相色谱串联质谱法检测黄花鱼中硫黄素T的方法。方法样品以甲醇作为萃取溶剂,经C18反相色谱柱分离,以甲醇和0.1%甲酸酸化的0.02mol/L乙酸铵作为流动相梯度洗脱,流速为0.30 mL/min,采用电喷雾离子源,正离子多反应检测扫描方式监测,基质匹配标准曲线法定量。结果硫黄素T在1~100 ng/mL范围内具有良好的线性,方法定量限为1.2μg/kg,回收率为85.5%~93.8%,相对标准偏差为0.5%~3.1%。结论该检测方法操作简单,结果准确,可应用于黄花鱼中硫黄素T的日常检测。
Objective To establish a method for the determination of thioflavin T in yellow croaker by high performance liquid chromatography-tandem mass spectrometry.Methods Samples were extracted with methanol as solvent,separated by C18 reverse phase chromatography with gradient elution using methanol and 0.1%formic acidified 0.02 mol/L ammonium acetate as the mobile phases.The flow rate was 0.30 mL/min.The electrospray ionization source was used.The positive ion multiple reaction detection scanning method was used for monitoring.The matrix matching standard curve method was used for quantitative analysis.Results The thioflavin T had good linearity in the range of 1-100 ng/mL.The limit of quantitation was 1.2μg/kg,the recoveries were 85.5%-93.8%,and the relative standard deviation were 0.5%-3.1%.Conclusion The method is simple and the result is accurate.It can be applied to the daily detection of thioflavin T in yellow croaker.
