摘要
【目的】本研究旨在探究猪CKM基因的多态性及其mRNA表达水平,通过对CKM基因起始密码子上游3 kb区域进行多态性检测,发现了单核苷酸多态性位点C-1185T和C-1324T,且该位点符合哈迪-温伯格定律(P<0.05)。【方法】利用实时荧光定量PCR技术对CKM基因在藏猪和大约克猪不同组织的mRNA表达量进行检测。【结果】发现在同一品种猪种,CKM基因的mRNA表达水平从高至低依次为背最长肌>背脂>肝脏组织,不同猪种内CKM基因在背最长肌、背脂和肝脏组织中存在差异表达,在背最长肌中,藏猪(TP)极显著高于大约克猪(YY)(P<0.01),在背脂中,大约克猪(YY)显著高于藏猪(TP)(P<0.05),在肝脏组织中,藏猪(TP)极显著高于大约克猪(YY)(P<0.01)。【结论】推测CKM基因与骨骼肌的生长发育呈负相关。本研究为下一步深入开展CKM基因功能研究奠定基础。
【Objective】The purpose of this study was to investigate the polymorphism of the pig CKM gene and its mRNA expression level.By detecting the polymorphism in the 3 kb region upstream of the start codon of the CKM gene,single nucleotide polymorphism sites C-1185T and C-1324T,and this site complies with Hardy-Weinberger's law(P<0.05).【Method】Real-time quantitative PCR was used to detect the mRNA expression of CKM gene in different tissues of Tibetan pigs and Yorkshire pigs.【Result】It was found that in the same breed of pig breeds,the mRNA expression level of CKM gene in order from the highest to the lowest was the longest back muscle>back fat>liver tis-sue,CKM genes in different pig breeds were differentially expressed in longissimus dorsi muscle,back fat and liver tissues.In longissimus dorsi muscle,Tibetan pig(TP)was significantly higher than Yorkshire pig(YY)(P<0.01),in back fat,Yorkshire pigs(YY)were significantly higher than Tibetan pigs(TP)(P<0.05),and in liver tissue,Tibetan pigs(TP)were extremely significantly higher than Yorkshire pigs(YY)(P<0.01).【Conclusion】It is speculated that there is a negative correlation between the CKM gene and the growth and development of skeletal muscle.This study lays the foundation for further research on the function of CKM gene.
