采用花粉管通道法遗传转化月季石榴的研究 被引量:4

Agrobacterium-mediated Transformation of Dwarf Pomegranate(Punica granatum)by Pollen Tube Pathway Method

摘要 以月季石榴为试验材料,结合花粉管通道法和农杆菌介导法进行浸染转化,探究了不同浸染介质及不同授粉方式对坐果率、出芽率、阳性率的影响。对授粉后花粉管的萌发进行观察,发现在授粉后24 h进行菌液浸染可能使菌液深入到花粉管,从而提高转化效率。对转化后的1819棵实生苗进行GUS染色和初步筛选,获得了82棵待检测植株;使用PCR扩增技术对转基因阳性植株进行鉴定,得到了14棵转PgLCBF1基因的月季石榴植株。对不同处理组合所获得的月季石榴的坐果率、出芽率、阳性率的研究结果表明:当浸染介质为5%蔗糖、授粉方式为授粉后24 h浸染时,月季石榴的遗传转化效果较好。 The agrobacterium-mediated transformation of dwarf pomegranate(Punica granatum L.var.nana)by pollen tube pathway method was carried out in the paper.The effects of different infecting media and different pollination ways on fruit setting rate,budding rate and transformed rate were studied.The germination of pollen tube after pollination was observed,and it was found that the infestation within 24 h after pollination might increase the transformation efficiency.The 1819 transformed plants were initially screened by GUS staining,and 82 plants were obtained.PCR amplification technology was used to identify transgenic plants,and 14 PgLCBF1-transgenic dwarf pomegranate plants were obtained.Calculating the fruit setting rate,budding rate and transformed rate of four different treatments,we found that the treatment(infection medium was MS+5%sucrose,transformed within 24 hours after pollination)was better for transformation.
