芪苈强心胶囊通过TLR4/NF-κB通路减轻大鼠心肌炎性反应及损伤 被引量:7

摘要 目的观察芪苈强心胶囊对心肌梗死(MI)大鼠心肌免疫炎性反应及结构损伤的影响及机制。方法2019年4—6月于宣城市人民医院病理科进行实验。将32只SD大鼠以随机数字表法分为假手术组、MI组、MI+芪苈强心组及MI+卡托普利组,各8只,采用冠状动脉结扎法建立大鼠MI模型。术后假手术组、MI组给予生理盐水10 ml·kg^(-1)·d^(-1),MI+芪苈强心组、MI+卡托普利组分别以芪苈强心胶囊1.2 g·kg^(-1)·d^(-1)及卡托普利25 mg·kg^(-1)·d^(-1),均每日1次,灌胃2周。采用Western-blot检测各组大鼠心肌Toll样受体4(TLR4)、核转录因子κB(NF-κB)、肿瘤坏死因子-α(TNF-α)、白介素^(-1)β(IL^(-1)β)蛋白表达,TUNEL染色检测各组心肌细胞凋亡,HE染色观察各组心肌形态学变化。结果与假手术组比较,MI组心肌TLR4、NF-κB、TNF-α、IL^(-1)β蛋白表达及心肌细胞凋亡率均显著升高;与MI组比较,MI+芪苈强心组上述指标均显著降低(t/P=7.536/0.002、6.103/0.004、6.660/0.003、6.655/0.003、5.548/0.005);MI+芪苈强心组与MI+卡托普利组上述指标比较差异无统计学意义(P均>0.05)。结论芪苈强心胶囊可能通过TLR4/NF-κB信号通路减轻心肌梗死大鼠心肌免疫炎性反应和结构损伤。 Objective To observe the effect and mechanism of Qiliqiangxin capsule on myocardial immune inflammatory response and structural damage in rats with myocardial infarction(MI).Methods The experiment was conducted in the Department of Pathology,Xuancheng People's Hospital from April to June 2019.Thirty-two rats were divided into sham operation group,MI group,MI+Qiliqiangxin capsule group and MI+captopril group by random number table method,each with 8 rats.The SD rat MI model was established by coronary artery ligation.Postoperative sham operation group and MI group were given normal saline 10 ml·kg^(-1)·d^(-1),MI+Qiliqiangxin capsule group and MI+captopril group were given Qiliqiangxin capsule 1.2 g·kg^(-1)·d,respectively^(-1) and captopril 25 mg·kg^(-1)·d^(-1),once a day,gavage for 2 weeks.Western blot was used to detect myocardial TLR4,NF-κB,TNF-α,IL^(-1)βprotein expression in each group of rats.TUNEL staining detected cardiomyocyte apoptosis in each group.The morphological changes of myocardium in each group were observed by HE staining.Results Compared with the sham operation group,the expression of myocardial TLR4,NF-κB,TNF-α,IL^(-1)βand the apoptosis rate of myocardial cells in the MI group were significantly increased;compared with the MI group,the above indicators of the MI+Qiliqiangxin capsule were significantly reduced(t/P=7.536/0.002,6.103/0.004,6.660/0.003,6.655/0.003,5.548/0.005);MI+Qiliqiangxin group and MI+captopril group has no significant difference(all P>0.05).Conclusion The Qiliqiangxin capsule may reduce myocardial immune inflammatory response and structural damage in rats with myocardial infarction through TLR4/NF-κB signaling pathway.
