摘要
为探究p38 MAPK抑制剂SB203580对热应激雌性雏鸡主要生殖激素分泌的影响,将72只1日龄文昌鸡母雏随机分为对照组(CK)、热应激组(HS)和抑制剂组(HS+I),从第三周开始,每天中午12∶00对HS、HS+I组雏鸡进行热应激2 h,每周龄末在热应激前30 min给HS+I组雏鸡腹腔注射SB203580(5 mg/kg体重)。采用ELISA对血浆中促性腺激素释放激素(GnRH)、促卵泡素(FSH)、促黄体生成素(LH)、雌二醇(E2)的含量进行测定。结果显示:HS组GnRH含量在3周龄时比CK组显著上升,而在4、5、6周龄时显著降低,HS+I组在3、4周龄时比HS组显著降低(P<0.05);HS组FSH含量在3周龄比CK组显著增加而在6周龄显著减少,HS+I组在3周龄比HS组显著减少(P<0.05);HS组LH含量在3周龄比CK组显著上升而在4、6周龄显著下降,HS+I组在3周龄比HS组显著减少(P<0.05);HS组E2含量在3周龄比CK组显著上升而在5、6周龄显著下降,HS+I组在3周龄比HS组显著减少(P<0.05)。结果表明:热应激扰乱血浆中GnRH、FSH、LH、E2的正常水平,SB203580可抑制热应激雏鸡体内GnRH、FSH、LH、E2的分泌。
Seventy-two 1-day-age female Wenchang chicks were randomly divided into the control group(CK),the heat stress group(HS)and the inhibitor group(HS+I).Starting from the third week,the chicks in HS and HS+I were subjected to heat stress for 2 hours at 12∶00(noon)every day.At the end of each week,p38 MAPK inhibitor,SB203580(5 mg/kg BW)was intraperitoneally injected into chicks in the HS+I 30 minutes before heat stress.The contents of GnRH,FSH,LH and E2 in plasma were determined by ELISA.GnRH in the HS increased significantly at the 3 rd week than the CK,and decreased significantly at other weeks,and which of HS+I was significantly lower than the HS at3 rd and 4 th week(P<0.05).FSH in the HS increased significantly at the 3 rd week and decreased significantly at the6 th week than the CK,the HS+I decreased significantly at the 3 rd week than the HS(P<0.05).LH in the HS increased significantly at the 3 rd week than the CK,but decreased significantly at the 4 th and 6 th week,and which of HS+I decreased significantly at the 3 rd week than the HS(P<0.05).The E2 in the HS increased significantly at the3 rd week than the CK,but decreased significantly at the 5 th and 6 th week,and which of the HS+I decreased significantly at the 3 rd week than the HS(P<0.05).The results indicated that heat stress disturbed the normal levels of GnRH,FSH,LH and E2 in plasma,and SB203580 inhibited the secretion of GnRH,FSH,LH and E2 in chicks under heat stress.
