哺乳仔猪粪便中四环素和头孢耐药菌的分离及鉴定

Isolation and Identification of Tetracycline and Cephalosporin-Resistant Bacteria from Suckling Piglets Faeces

摘要 笔者通过对哺乳仔猪粪便中耐药菌进行分离及鉴定,来研究有关抗生素类化学药物滥用所引发的耐药菌株大量出现问题。试验以哺乳仔猪为研究对象,制备含有四环素和头孢的抗性平板,通过微生物的纯培养技术对粪便中四环素耐药细菌以及头孢耐药细菌进行分离纯化,再对筛选纯化后的2种抗性细菌进行DNA提取,并用16srDNA引物进行PCR扩增测序和分子生物学鉴定。结果四环素耐药菌数量为6.000×107CFU/g,抗性细菌所占的比例为0.21%;头孢氨苄耐药细菌数量为1.026×107CFU/g,抗性细菌所占的比例为37.34%。四环素和头孢耐药细菌均为大肠埃希氏菌(Escherichiacoli)。本研究为合理使用四环素类和头孢类抗生素提供数据支撑。 The isolation and identification of drug-resistant bacteria in the feces of Suckling piglets were used to study the problems of large Numbers of drug-resistant strains caused by the abuse of antibiotic chemical drugs.This experiment by lactation piglet as the research object,preparation containing tetracycline and cephalosporin resistant plate,through the pure culture of microorganisms technology on faeces tetracycline resistant bacteria and cephalosporin resistant bacteria in separation and purification,again after the screening and purification of two kinds of resistant bacteria for DNA extraction,and 16s rDNA sequencing primers for PCR amplification and molecular biology identification.The results showed that the number of tetracycline resistant bacteria was 6.000×107 CFU•g-1,and the proportion of resistant bacteria was 0.21%.The number of cefalexin resistant bacteria was 1.026×1010 CFU•g-1,and the proportion of resistant bacteria was 37.34%.Both tetracycline and cefore-resistant bacteria were Escherichia coli.This study provides data support for the rational use of tetracycline and cephalosporin antibiotics.
