摘要
目的:建立HPLC法测定肠外营养液(TNA)中维生素C(Vc)注射液含量的方法。方法:采用Zirchrom Kromasil C18(250 mm×4.6 mm,5μm)色谱柱;流动相:0.34%(w/v)磷酸二氢钾-氢氧化四丁基铵-乙腈(92.5∶2.5∶5,v/v/v,pH 3.0);流速:0.8 ml/min;检测波长:210 nm;柱温:30℃。结果:线性回归方程:Y=27105 X-10325(r=0.9998),线性范围为:50~600μg/ml;平均回收率分别为99.23%,RSD为1.06%;重复性RSD为1.78%。结论:该方法操作简单,快速准确,且分离度和重复性均较好,可用于成分复杂的TNA中Vc注射液的含量测定,同时也为进一步深入研究TNA中加入适宜浓度Vc注射液的可行性奠定了基础。
Objective:To establish a high-performance liquid chromatography(HPLC)method for the determination of vitamin C(Vc)injection in total parenteral nutrition admixture(TNA).Methods:Zirchrom Kromasil C18(250 mm×4.6 mm,5μm)chromatographic column;Flow phase:0.34%(w/v)potassium dihydrogen phosphate/hydroxide tetrabutylammonium/acetonitrile(92.5,2.5,5,v/v/v,pH 3.0);Flow rate:0.8 ml/min;Detection wavelength:210 nm;Column temperature:30°C.Results:Linear regression equation:Y=27105X-10325(r=0.9998),linear range:50~600μg/ml;The average recovery rate was 99.23%(RSD 1.06%);Repetition RSD was 1.78%.Conclusion:The method is simple,fast,and accurate and has good separation and repeatability.It can be used for the determination of Vc injection in complex TNA.At the same time,it also laid a foundation for further study on the feasibility of adding an appropriate concentration of Vc injection into TNA.
