基于网络药理学探究水陆二仙丹治疗膜性肾病作用机制 被引量:7

Exploration of the Mechanism of Shuilu Erxian Dan in the Treatment of Membranous Nephropathy Based on Network Pharmacology

摘要 目的应用网络药理学探究水陆二仙丹的有效成分与作用机制。方法基于TCMSP数据库,筛选水陆二仙丹的活性成分和作用靶点,借助GeneCards与OMIM数据库筛选水陆二仙丹与膜性肾病相关的靶点,利用Cytoscape 3.7.1构建药物调控网络,得到疾病基因靶点与药物靶点汇总结果。利用STRING数据库构建水陆二仙丹治疗膜性肾病的蛋白互作网络。运用R3.6.1进行GO与KEGG基因功能富集分析。结果筛选出9种水陆二仙丹中的生物活性成分,其中金樱子7种、芡实2种。共筛选出药物潜在作用靶点1308个,其中金樱子711个、芡实597个,最终获得56个与膜性肾病相关的作用靶点。PPI网络中共包括63种功能蛋白,其中存在454个蛋白质相互作用关系。GO共富集89个注释条目,KEGG共富集115个注释条目。结论运用网络药理学分析方法,探究了水陆二仙丹治疗膜性肾病的物质基础与作用机制,发现水陆二仙丹中含有槲皮苷、山柰酚、谷甾醇、维生素E等成分作用于VEGFA、AR、CASP3、CASP8等多个靶点和肿瘤坏死因子信号通路、AGE-RAGE信号通路、TNF信号通路等多条通路,体现出其多成分、多靶点、多通路的整合调节作用特点。为今后深入探究水陆二仙丹治疗膜性肾病的有效作用成分与分子机制研究提供了理论基础。 Objective To explore the effective components and mechanism of Shuilu Erxian Dan by using network pharmacology.Methods Based on the TCMSP database,the active ingredients and target of Shuilu Erxian Dan(euryurum-cherruria chinensis)were screened,and the targets related to membranous nephropathy were screened with the help of GeneCards and OMIM databases.Cytoscape3.7.1 was used to build a drug regulatory network,and the summary results of disease gene targets and drug targets were obtained.A protein interaction network for the treatment of membranous nephropathy was constructed by using STRING database.R3.6.1 was used for functional enrichment analysis of GO and KEGG genes.Results In this study,9 kinds of effective ingredients were screened out,including 7 kinds of Rosa laevigata and 2 kinds of Euryale ferox-Rosa laevigata.A total of 1308 potential drug targets were screened out,including 711 cherry seeds and 597 Euryale ferox-Rosa laevigata seeds,and 56 targets related to membranous nephropathy were finally obtained.PPI network includes 63 functional proteins,among which 454 proteins interact with each other.GO enriched 89 annotation items in total,and KEGG enriched 115 annotation items in total,covering multiple cell signaling pathways.Conclusion This study using network pharmacology analysis method,explores the Shuilu Erxian Dan material basis for the treatment of membranous nephropathy and the mechanism of action,Shuilu Erxian Dan was found to contain multiple components acting on multiple targets and pathways,and the known pharmacological effects reported in literatures are consistent,reflects its composition,the integration of multiple targets,multiple pathways regulating role.It provides a theoretical basis for further research on the effective components and molecular mechanism of Shuilu Erxian Dan in the treatment of membranous nephropathy.
