萝卜素影响下化疗药物对胃癌细胞增殖的影响

摘要 目的:探讨萝卜硫素(sulforaphane,SFN)是否影响DNA损伤药物5-氟尿嘧啶(5-fluorouracil,5-FU)和顺铂(cisplatin)对胃癌细胞增殖的抑制效应及其机制。方法:体外培养胃癌SGC7901细胞,采用CCK8法检测细胞的活力,Western Blot分析SGC7901细胞中WEE1蛋白表达,采用pcDFNA-WEE1质粒过表达技术对SGC7901细胞中WEE1蛋白进行过表达。结果:萝卜硫素能明显增强5-氟尿嘧啶和顺铂对胃癌SGC7901细胞增殖的抑制效应,萝卜硫素还呈浓度、时间依赖性的抑制SGC7901细胞中WEE1蛋白表达;pcDFNA-WEE1质粒转染能明显上调SGC7901细胞中WEE1蛋白表达,并且显著降低了萝卜硫素联合DNA损伤药物对胃癌SGC7901细胞增殖的抑制效应。结论:萝卜硫素可通过抑制WEE1激酶表达增加胃癌细胞对DNA损伤药物的敏感性。 Objective: To discuss whether sulforaphane could affect the inhibitory effects of DNA damage drugs 5-fluorouracil(5-FU) and cisplatin on the proliferation of gastric cancer cells. Methods: Gastric cancer SGC7901 cells were cultured in vitro, CCK8 method was adopted to detect cellular viability, Western blot to analyze the expressions of WEE1 protein in SGC7901 cells, pcDFNA-WEE1 plasmid overexpression technology was used to measure the overexpression of WEE1 protein in SGC7901 cells. Results: Sulforaphane could enhance the inhibitory effects of 5-FU and cisplatin on the proliferation of gastric cancer cells notably, it could inhibit the expressions of WEE1 protein in SGC7901 cells in a dose-and time-dependent manner;pcDFNA-WEE1 plasmid transfection could notably upregulate the expressions of WEE1 protein in SGC7901 cells, and it could notably reduce the inhibitory effects of sulforaphane and DNA damage drugs on the proliferation of gastric cancer SGC7901 cells. Conclusion:Sulforaphane could increase the sensitivity of gastric cancer cells to DNA damage drugs through inhibiting the expressions of WEE1 kinase.
