摘要
目的探究GNAI2在食管鳞癌组织中的表达及其临床意义。方法选取50例手术切除的食管鳞癌患者癌组织及与其配对癌旁组织样本,用免疫组织化学法检测组织样本中GNAI2蛋白的表达,并分析GNAI2蛋白与食管鳞癌患者临床病理特征的相关性;COX回归分析各因素对食管鳞癌患者预后的影响;Kaplan-Meier法绘制生存曲线,并通过Log-rank检验GNAI2与生存时间之间的相关性。结果食管鳞癌及癌旁组织中均可见GNAI2蛋白阳性表达,为细胞质出现棕黄色颗粒。癌组织中GNAI2免疫组化学评分显著高于癌旁组织;且随着癌组织分化程度的降低,GNAI2免疫组化评分逐渐升高。GNAI2蛋白与食管鳞癌分化程度、侵袭深度、淋巴结转移、TNM分期和5年生存情况具有相关性,与患者的性别、年龄不具有相关性。分化程度及GNAI2蛋白的表达为食管鳞癌患者预后的独立危险因素。GNAI2蛋白表达阳性组5年生存率(18.75%)明显低于表达阴性组(55.56%)。结论GNAI2蛋白高表达可能促进了食管鳞癌的发生、发展,抑制GNAI2的表达可能是食管鳞癌靶向治疗的潜在靶点之一。
Objective To explore the expression and clinical significance of GNAI2 in esophageal squamous cell carcinoma.Methods Fifty samples of cancerous tissue and its adjacent tissue samples were collected from patients by resection,and the expression of GNAI2 was detected by immunohistochemical method.Then,the correlations between expression and clinicopathologic features,and between the expression and the 5-year survival prognosis of esophageal squamous cell carcinoma were analyzed.In details,COX regression analysis method was applied to draw the impact of various factors on the prognosis of esophageal squamous cell carcinoma;survival curves were pictured by Kaplan-Meier method,after which the correlation between GNAI2 and survival time was tested via Log-rank method.Results The positive expression of GNAI2 protein can be found both in esophageal squamous cell carcinoma and adjacent tissues,and the brown granules appeared in the cytoplasm.The immunohistochemical score of GNAI2 in cancer tissues was significantly greater than that in paracancerous tissues,and the GNAI2 immunohistochemical score increased gradually with the decreasing of the degree of differentiation of cancer tissues.GNAI2 protein was significantly correlated with the degree of differentiation,depth of invasion,lymph node metastasis,TNM stage and 5-year survival of esophageal squamous cell carcinoma,but not with sex and age.The degree of differentiation and the expression of GNAI2 protein were independent risk factors for prognosis in patients with esophageal squamous cell carcinoma.The 5-year survival rate of GNAI2 protein positive group(18.75%)was significantly lower than that of negative expression group(55.56%).Conclusion The high expression of GNAI2 protein may promote the occurrence and development of esophageal squamous cell carcinoma.And inhibiting the expression of GNAI2 may be one of the potential targets for targeted therapy of esophageal squamous cell carcinoma.
