中药提取物对痤疮丙酸杆菌诱导THP-1细胞炎症因子表达的抑制作用 被引量:17

摘要 目的探究痤疮丙酸杆菌及其外膜囊泡对痤疮炎症的作用,筛选中药提取物,找到一种可以抑制痤疮丙酸杆菌生长同时抑制其外膜囊泡释放起到抑制痤疮炎症的组合。方法采用牛津杯法测量抑菌圈直径、测定最小抑菌浓度判断中药提取物的抑菌活性,采用BCA蛋白浓度测定方法检测中药提取物对细菌释放外膜囊泡浓度的影响,并用ELISA试剂盒检测中药提取物对痤疮丙酸杆菌诱导THP-1细胞炎症因子表达的抑制作用。结果苦参乙醇提取物具有较好的抑菌活性,对痤疮丙酸杆菌的抑菌圈为(14.35±0.64)mm,最小抑菌浓度为0.20 mg/mL。艾叶乙醇提取物对外膜囊泡释放具有抑制作用,最小抑制浓度为10μg/mL。苦参乙醇提取物、艾叶乙醇提取物的组合较好的抑制了痤疮丙酸杆菌诱导的THP-1细胞炎症因子TNF-α、IL-1β、IL-8表达。结论苦参乙醇提取物和艾叶乙醇提取物的组合具有抑制痤疮的效果。 AIM To explore the effects of Propionibacterium acnes and its outer membrane vesicles(OMVs)on acne inflammation.To screen several extracts of Chinese herbal medicines,and to find their appropriate combinative use to suppress P.acnes growth while reducing PEVs release to alleviate acne occurence.METHODS The diameter of the inhibition zone was measured by the Oxford Cup test,and the minimum inhibitory concentration was determined to evaluate the antibacterial activity of extracts of several Chinese herbal medicines.The effects of Chinese herbal medicine extracts on the release of OMVs were determined by BCA method.And the extracts’inhibitory effects on the expression of inflammatory cytokines induced by P.acnes were detected with ELISA kits.RESULTS The ethanol extract of Sophora flavescens demonstrated good antibacterial activity with its inhibition zone of(14.35±0.64)mm and minimum inhibitory concentration of 0.20 mg/mL.The ethanol extract of Artemisia argyi displayed an inhibitory effect on the release of OMVs with a minimum inhibitory concentration of 10μg/mL.The combinative use of ethanol extracts of Sophora flavescens and Artemisia argyi suppressed the expression of inflammatory cytokines of TNF-α,IL-1βand IL-8 of THP-1 cells triggered by P.acnes.CONCLUSION The combinative use of ethanol extracts of Sophora flavescens and Artemisia argyi has an effect against acne.
