细胞增殖上调因子促进神经胶质瘤细胞增殖的潜在机制 被引量:2

摘要 目的探讨细胞增殖上调因子(URGCP)对神经胶质瘤细胞增殖的影响及潜在机制。方法采用Western印迹检测URGCP在正常星形胶质NHA细胞和神经胶质瘤A172、U251和U87细胞中的表达。利用脂质体2000分别转染URGCP-siRNA和pcDNA3.0-URGCP重组质粒以构建沉默URGCP的U87细胞和过表达URGCP的A172细胞。MTT法和克隆形成实验检测URGCP对U87细胞和A172细胞增殖的影响,Western印迹检测URGCP对细胞周期蛋白(Cyclin)D1和CyclinE1蛋白及核转录因子(NF)-κB信号通路相关蛋白IκBα、p-IκBα、NF-κB/p65表达的影响。结果URGCP在神经胶质瘤A172、U251和U87细胞中的表达水平依次增大,且均明显大于正常星形胶质NHA细胞(P<0.05)。URGCP-siRNA使U87细胞活力减弱,细胞克隆数减少,CyclinD1、CyclinE1、p-IκBα和胞核内NF-κB/p65蛋白表达下降,差异有统计学意义(P<0.05),胞质内NF-κB/p65表达升高;pcDNA3.0-URGCP使A172细胞细胞活力增强,细胞克隆数增多,CyclinD1、CyclinE1、p-IκBα和胞核内NF-κB/p65蛋白表达升高,差异有统计学意义(P<0.05),胞质内NF-κB/p65表达下降,差异有统计学意义(P<0.05)。URGCP-siRNA和pcDNA3.0-URGCP均未影响IκBα蛋白的表达。结论URGCP在神经胶质瘤细胞中表达升高,可通过激活NF-κB信号通路促进癌细胞的增殖。
