摘要
目的探讨多聚腺苷二磷酸核糖聚合酶1(PARP-1)表达对前列腺癌细胞顺铂(DDP)耐药性的影响。方法采用转染PARP-1 siRNA抑制前列腺癌细胞中PARP-1的表达,RT-PCR和Western blot检测细胞中PARP-1以及凋亡信号通路相关蛋白的表达差异,CCK-8检测耐药指数,计算DDP IC50。结果 PARP-1蛋白在耐药细胞PC-3/DDP中的表达量明显高于亲本细胞PC-3,两组比较差异有统计学意义(P<0.01)。下调PARP-1表达可增强前列腺癌细胞的DDP敏感性,逆转耐药细胞的耐药性。信号通路相关蛋白Bcl-2和pERK显著下调。结论 PARP-1在前列腺癌细胞表达水平与细胞对DDP耐药性密切相关,PARP-1表达抑制可以增强DDP对前列腺癌细胞的敏感性,其分子机制可能是通过激活凋亡通路抑制ERK磷酸化。
Objective To investigate the effects of poly(ADP-ribose)polymerase-1(PARP-1)on cisplatin resistance of prostate cancer cells.Methods The expression of PARP-1 was down regulated by PARP-1 siRNA.The protein levels of PARP-1,Bcl-1,Bax,ERK and pERK were detected by Western blot or RT-PCR.The cell viability and IC50 of cisplatin were deteceted by the CCK-8 assay.Results The expression of PARP-1 protein was increased markedly in PC-3/DDP vs.PC-3 cells(P<0.01).Reduced PARP-1 expression could enhance the sensitivity to cisplatin in PC-3 cells,rescue the resistance of cisplatin in PC-3/DDP,and downregulate the protein levels of Bcl-2 and pERK.Conclusions Expression of the PARP-1 protein in prostate cancer cells is related to cellular sensitivity to cisplatin.PARP-1 siRNA treatment could increases the sensitivity of prostate cancer cell to cisplatin.The mechanism may be promote inhibit apoptosis pathway and phosphorylation of ERK.
