摘要
为探究大河乌猪MSTN基因第3外显子的多态性,分析其是否能作为调控眼肌面积的主效基因或分子标记位点,为大河乌猪提高瘦肉率的选育提供理论依据,采用PCR-Taq I-RFLP技术探究大河乌猪MSTN基因第3外显子的多态性,并将基因型与眼肌面积进行关联分析,筛选大河乌猪眼肌面积的高产基因型。结果显示:大河乌猪MSTN基因第3外显子酶切后,得到2种基因型,一种含有六个条带(439、315、287、152、124、28 bp)的CTGA基因型,另一种含有四个条带(287、152、124、28 bp)的CTGG基因型。将2种基因型与眼肌面积进行关联分析后,得到CTGA基因型调控的眼肌面积为27.23 cm^2,CTGG基因型调控的眼肌面积为31.60 cm^2,大河乌猪CTGG基因型调控的眼肌面积极显著高于CTGA基因型(P<0.01)。试验结果表明,MSTN基因第3外显子CTGG基因型有效地提高了大河乌猪的眼肌面积,可作为影响大河乌猪眼肌面积的候选基因或分子标记位点。
To explore the polymorphism of exon 3 of MSTN gene in Dahewu pig,and analyze whether it can be used as a major gene or molecular marker site to regulate the eye muscle area,and provide a theoretical basis for the selection of Dahewu pig to improve the lean meat rate,in this study,the polymorphism of exon 3 of the MSTN gene of 79 Dahewu pigs was investigated by using PCR-TaqI-RFLP technique,and the genotype was correlated with the eye muscle area,so as to screen the genotype of the eye muscle area of Dahewu pig with high yield.The results showed that after exon 3 of MSTN gene was digested,two genotypes were obtained,one was CTGA with six bands(439,315,287,152,124,28 bp),the other was CTGG with four bands(287,152,124,28 bp).The correlation analysis between the two genotypes and the eye muscle area showed that the eye muscle area regulated by the CTGA genotype was 27.23 cm^2,and the eye muscle area regulated by the CTGG genotype was 31.60 cm^2.The area of the eye muscle regulated by CTGG genotype was significantly higher than that of CTGA genotype(P<0.01).The results showed that the CTGG genotype of the third exon of MSTN gene could effectively improve the eye muscle area of Daheuwu pig,and could be used as the main gene or molecular marker site of influencing the eye muscle area of Daheuwu pig.
