高效液相色谱测定粮食中L-色氨酸的方法分析 被引量:5

摘要 运用超高效液相色谱建立粮食中L-色氨酸含量的检测方法。样品经过6 mol/L氢氧化钠和5%氯化亚锡配成的水解保护液,在110℃烘箱下水解20 h,经Dikmatech C18色谱柱(Endeavorsil 1.8μm 50 mm×2.1 mm),磷酸氢二钠缓冲液(pH3.0)为流动相等梯度洗脱,DAD检测器278 nm,在0.728~14.56μg/mL范围内线性良好。标准曲线方程为Y=9.531X—1.0395,相关系数R2=0.9997,L-色氨酸最低检出限为0.006%,最低定量限为0.019%。粮食中如稻谷和玉米,L-色氨酸含量小于0.1%,该方法精密度≤10%;粮食中如小麦和大豆,L-色氨酸含量高于0.1%时,该方法精密度≤3%,加标回收率在90.07%~96.27%。该方法检测时间短,准确可靠,适用于粮食中色氨酸的定量检测。 A method for the determination of L-tryptophan in grain was established by ultra high performance liquid chromatography.Samples after 6 mol/L NaOH and 5%match hydrolysis protection of SnCl2 hydrolysis of 20 h under 110℃oven,via Dikmatech C18 chromatographic column(Endeavorsil 1.8μm 50 mm×2.1 mm),Na2HPO4 buffer(pH3.0)as equal flow gradient elution,DAD detector,278 nm in 0.728~14.56μg/mL range linear good.Standard curve equation for Y=9.531X—1.0395,R2=0.9997,the minimum detection limit of L-tryptophan was 0.006%,and the minimum quantitative limit was 0.019%.When the content of L-tryptophan in grains such as rice and maize is less than 0.1%,the precision of this method is less than or equal to 10%;when the content of L-tryptophan in grains such as wheat and soybean is higher than 0.1%,the precision of this method is less than or equal to 3%,and the standard recovery rate is between 90.07%and 96.27%.The method is accurate and reliable,and is suitable for the quantitative determination of tryptophan in grain.
