摘要
目的运用网络药理学方法对灵归方进行研究,探究灵归方治疗少弱精子症的可能机制。方法利用TCMSP、BATMAN-TCM及相关文献资料检索灵归方的有效成分和作用靶点,借助OMIM和Genecards数据库检索少弱精子症的相关靶点,运用韦恩图工具获取交集靶点并导入String数据库制作蛋白互作网络,获取核心靶点,利用Cytoscape软件将疾病-药物-有效成分-靶点网络可视化处理。在DAVID数据库中对核心靶点进行GO和KEGG富集分析。结果研究发现槲皮素、山奈酚可能为灵归方的关键成分,AR与ACh E可能是治疗过程中最重要的靶点,PI3K/Akt、AGE-RAGE、MAPK、Fox O、JAK/STAT、m TOR、Wnt等信号通路是灵归方治疗少弱精子症的重要通路。结论利用网络药理学的方法研究灵归方的有效成分、靶点和通路,体现了灵归方多成分、多靶点、多通路的治疗特点,为进一步研究灵归方治疗少弱精子症的相关机制提供了新方向和新思路。
Objective To investigate the possible mechanism of Linggui Prescription in the treatment of oligoasthenozoospermia by using network pharmacology.Methods TCMSP,BATMAN-TCM and related literature were used to collect the active ingredients and targets of Linggui Prescription.Relevant targets for oligoasthenozoospermia were retrieved using OMIM and Genecards databases.The intersecting targets were obtained by using the Wayne diagram tool and imported into the string database to make protein interaction network and obtain the core targets.The disease-drug-active ingredient-target network was visualized by Cytoscape software.GO and KEGG enrichment analysis was performed on the core targets in the DAVID database.Results Quercetin and kaempferol may be the key components of Linggui Prescription,AR and ACh E may be the most important targets in the treatment process,and PI3 K/Akt,AGE-RAGE,MAPK,Fox O,JAK/STAT,m TOR,Wnt and other signaling pathways are important pathways of Linggui Formula in the treatment of oligoasthenozoospermia.Conclusion In this study,the active ingredients,targets and pathways of Linggui Prescription were preliminarily studied using the method of network pharmacology.The results of the study reflected the treatment characteristics of multi-component,multi-target and multi-pathway of Linggui Prescription,and provided a new direction and new ideas for further study of the related mechanism of Linggui Prescription in the treatment of oligoasthenozoospermia.
