鸭坦布苏病毒JJZZ1801株分离鉴定及其E蛋白序列分析 被引量:3

Isolation and Identification of Duck Tembusu Virus JZ1801 Strain and Sequence Analysis of Viral Envelope Protein

摘要 为了解鸭群发病原因,对疑似鸭坦布苏病毒(DTMUV)感染的病鸭进行剖检,采集病变组织进行特异性RT-PCR检测和病原分离鉴定。结果显示:该鸭群为DTMUV感染,从病鸭脑组织分离到一株DTMUV,命名为JZ1801;基因序列分析显示,JZ1801的E基因与泰国分离株DK/TH/CU-1的E基因同源性最高(核苷酸相似度为97.8%),在系统进化树中处于一个新的亚分支,而与我国早期DTMUV分离株同源性相对较低,与之处于不同的亚分支;氨基酸位点差异分析显示,JZ1801的E蛋白氨基酸序列与我国早期分离株FX2010和BYD-1的E蛋白均具有9个差异位点,与疫苗株FX2010-180P的E蛋白存在15个氨基酸位点差异。结果表明近年来我国鸭群中流行的DTMUV E蛋白已经发生了一定程度的变异。 In order to determine the cause for the diseased ducks, the suspected duck Tembusu virus(DTMUV)infected ducks were necropsied, and the pathological tissues were sampled for specific RT-PCR and pathogen identification.The results showed that DTMUV infection was confirmed, and one strain of DTMUV was successfully isolated from the brain tissue and named JZ1801. Sequence analysis showed that E gene of JZ1801 exhibited the highest nucleotide homology(97.8%) with that of DK/TH/CU-1 isolated from Thailand and they were classified into a same new sub-branch in phylogenetic tree. But E gene of JZ1801 showed low nucleotide homology with the DTMUV strains isolated in early years in China, which were classified into different sub-branch. And there were 9 amino acids in E protein of JZ1801 different with FX2010 and BYD-1, and 15 amino acids different with the vaccine strain FX2010-180 P. These data indicated that E protein of DTMUV from the ducks circulated in China had obviously mutant in recent years.
