“胡椒+荜茇”挥发油化学成分分析及血脑屏障通透性研究 被引量:4

Study on the Chemical Composition of Volatile Oils from“Pepper and Fructus Piperis Longi”that Cross the Blood-Brain Barrier

摘要 目的探讨“胡椒+荜茇”挥发油化学成分及其血脑屏障通透性。方法采用水蒸气蒸馏法对胡椒、荜茇药对进行提取,得到的挥发油加入适量无水硫酸钠干燥,密闭低温保存。SD大鼠灌胃给药后取脑组织、脑脊液制备生物样品。用气相色谱-质谱联用(GC-MS)的方法,分别测定核心方剂中挥发油的成分、SD大鼠脑组织匀浆和脑脊液中的成分以及灌胃SD大鼠脑组织匀浆和脑脊液中的成分,对比分析核心方剂中透过血脑屏障的挥发油类成分。结果提取得到的药对挥发油为淡黄色透明油状液体,其中棕榈酸TMS衍生物可透过血脑屏障。结论“胡椒+荜茇”挥发油中棕榈酸TMS衍生物可透过血脑屏障,可能与脑卒中的治疗有关。 Objective To detect the volatile oil components that cross the blood-brain barrier in the treatment of stroke by pepper and fructus piperis longi.Methods Piper nigrum and Piper longum were extracted by steam distillation.The volatile oil was dried with anhydrous sodium sulfate and stored in airtight low temperature.The brain tissue and cerebrospinal fluid of SD rats were taken to prepare biological samples.The components of volatile oil in the core prescription,brain tissue homogenate and cerebrospinal fluid of SD rats,and the components of brain tissue homogenate and cerebrospinal fluid of SD rats were determined by gas chromatography-mass spectrometry(GC-MS).Results The volatile oil extracted in drug pair was a light yellow transparent oily liquid,and among them,the palmitate TMS derivative could pass through the blood-brain barrier.Conclusion The palmitate TMS derivative in volatile oil can pass through the blood-brain barrier,which may be closely related to the treatment of stroke.
