梗膏酶处理工艺参数的优化 被引量:2

Optimization of Enymolysis Conditions for Stem Paste

摘要 为提升再造烟叶品质,改善梗膏抽吸品质,使用木瓜蛋白酶对梗膏进行处理,采用正交试验方法优选出最佳工艺参数,对优选处理后的梗膏进行多酚氧化酶处理,选出最佳工艺参数。结果表明,经优化梗膏酶处理最优条件为:木瓜蛋白酶活力浓度3000 U/m L,酶解时间60 min,酶解温度55℃;多酚氧化酶活力浓度300 U/m L,酶解时间120 min,酶解温度55℃。经验证在该条件下处理的梗膏氨基酸态氮含量2.25%,制备的再造烟叶产品感官品质得到明显改善,说明优化结果合理可靠。 In order to improve the quality of reconstituted tobacco and the sensory quality of stem paste,stem paste was treated with papain,and the optimal process parameters were optimized by orthogonal test.The preferred treated stem paste was treated with polyphenol oxidase,and the optimal process parameters were selected.The results showed that the optimal condition for the optimization of the enzyme treatment was papain concentration of 3000 U/m L,enzymatic hydrolysis time of 60 min,and enzymatic hydrolysis temperature of 55℃;Polyphenol oxidase activity concentration of 300 U/m L,enzymatic hydrolysis time of 120 min,and the enzymatic hydrolysis temperature was 55℃.It was proved that the content of amino acid nitrogen in stem paste was 2.25%,and the sensory quality of the prepared reconstituted tobacco product was obviously improved,which indicated that the optimized results were reasonable and reliable.
