摘要
本文以13一甲基小檗碱为内标,用正相高效液相色谱一荧光检测的方法定量分析尿液中的小檗碱。考察了健康人一次口服0.2g盐酸黄连素糖衣片后96h内小檗碱尿药排泄量变化,按矩量法求得其体内药物平均滞留时间(MRT)为32.63 ±6. 15 h,累积尿药排泄量为 27.49 ± 4.74 u g;首次检测到小檗碱在人体内有四个未知的代谢峰。
A normal phase liquid chromatographic method (flurorescent detector) for determinating berberine (BER) concentration in human urine was reported. The 13-methylberberine was used as the internal standard. A mixture of dichloromethane - methanoldiethyl amine-acetic acid (90: 9:0:4:0.5) was served as the mobile phase. The dynamic change of excretion of BER in urine after oral administration of 0.2 g BER.HCL tablets in single dosage was observed and MRT and AUC was calculed to be 32.63 ± 6. 15 h and 27.49 ± 4.74 u g by statistical moment theory. Furthermore there were four metabolites of berberine chloride in human urine were detected for the first time.
