摘要
利用RT-PCR检测体系对贵州毕节烟区的烟草花叶病毒(tobacco mosaic virus,TMV)、黄瓜花叶病毒(cucumber mosaic virus,CMV)和马铃薯Y病毒(potato virus Y,PVY)3种病毒进行病害检测和株系鉴定,样品检测结果显示,TMV发生最严重,检出率为64.13%,其次是PVY,检出率为40.22%,CMV发生较轻,检出率为31.52%.通过同源性比较和构建系统进化树,结果表明,毕节烟区CMV均属于ⅠB亚组,TMV均属于种群Ⅰ,PVY均属于PVY N∶O株系,各株系间没有明显的地域差异,烟田周边马铃薯上分离物MQG1和MQG2,与烤烟上CMV,TMV和PVY 3种病毒的分离物亲缘关系较近,由此可以推测,烟田周边的马铃薯可能是烟田这3种病毒的主要毒源.
In this study,RT-PCR detection system was used for disease detection and strain identification of CMV,TMV and PVY viruses in the tobacco-growing areas in Bijie of Guizhou Province.The results of sample detection showed that the disease caused by TMV was most prevalent,with a detection rate of 64.13%.In contrast,the detection rates of PVY and CMV were 40.22%and 31.52%,respectively.The results of strain analysis showed that all the CMV isolates from the Bijie tobacco-growing area belonged to SubgroupⅠB,TMV isolates to PopulationⅠand PVY isolates to PVY N∶O strain,and there was no obvious regional difference among the strains.MQG1 and MQG2,the isolates from potato plants adjacent to the tobacco fields,were found to be close to the isolates of CMV,TMV and PVY on tobacco.It is,therefore,speculated that the potatoes around the tobacco field may be the main source of the three viruses.
