摘要
以普通三月茄为材料,利用农杆菌介导法对茄子进行遗传转化.通过设置一系列对照组,筛选出最佳诱导分化培养基及高效的抗生素组合、对比茄子子叶与下胚轴的诱导分化效果,从而对茄子的遗传转化体系进行优化和改良.结果表明:最佳诱导培养基为MS+6-BA 2 mg/L+ZT 1 mg/L;使用子叶作为外植体时,农杆菌介导的茄子遗传转化率达到了11.02%,其愈伤组织和不定芽的诱导率均显著高于下胚轴;当采用Carb 250 mg/L+Kan 100 mg/L的组合时,可以得到最佳的诱导效果,且在一定范围内,适当降低Carb与Kan浓度的比值,可以提高出愈率.
Eggplant variety“Sanyueqie”(March eggplant)was selected as the material of this experiment.The Agrobacterium-mediation method was used as a means of genetic transformation of the eggplant.Optimum differentiation medium and efficient antibiotic combinations were screened out and the differentiation results of the explants(cotyledon and hypocotyl of eggplant)were recorded in order to optimize and improve the genetic transformation system of eggplant by setting up a series of control groups.The results showed that the best induction medium was MS+6-BA 2 mg/L+ZT 1 mg/L.Agrobacterium-mediated genetic transformation of eggplant reached 11.02%when the cotyledon of eggplant was used as the explant.The induction rate of calli and adventitious buds was significantly higher than that of the hypocotyl.The best induction effect was obtained with the antibiotic combination of Carb 250 mg/L+Kan 100 mg/L.the callus-inducing rate was increased within a certain range by lowering the ratio of Carb/Kan concentration appropriately.
