干旱胁迫下白沙蒿SIP 2-1的表达和序列分析 被引量:1

Analysis of Sequence and Expression of SIP 2-1 in Artemisia sphaerocephala under Drought Stress

摘要 水通道蛋白家族(Aquaporins)的成员在植物体中起着膜通道的作用。基于转录组测序,对在干旱胁迫下呈显著下调表达的白沙蒿水通道蛋白基因AsSIP 2-1进行生物信息学分析。结果显示:AsSIP 2-1的cDNA序列的开放阅读框全长1 191 bp,共编码235个氨基酸;属于水通道蛋白SIP2-1家族的成员;AsSIP 2-1与5个参考物种的比对结果显示6个物种的总体同源性为82.92%,并具有SIP2-1家族的特有的NPL和NPA保守性结构域;AsSIP 2-1的等电点为9.32;分子量为26.145 kDa;分子式为C1218H1893N301O317S10;属于疏水性、稳定性蛋白;具有内质网膜保留信号EKKT基序;具有6个跨膜区,属于跨膜蛋白;无信号肽,属于非分泌蛋白。在干旱迫下,AsSIP 2-1表达量显著下调,参与了应答反应。 Aquaporins constitute water trans-membrane channels in plant cell.Based on the data of transcriptomic sequencing,the aquaporin gene AsSIP 2-1 in Artemisia sphaerocephala was analyzed through bioinformatics in the paper.The results showed that the AsSIP 2-1 was a member of the SIP2-1 family of aquaporins,whose cDNA contained 1191 bp open reading frame(ORF)and encoded 235 amino acids.The alignment of AsSIP2-1 with 5 reference species revealed that the 6 species were 82.92%in sequence similarity and all of them had the unique NPL and NPA domains of the SIP2-1 family.The analysis indicated that AsSIP2-1 was a hydrophobic and stable protein,whose isoelectric point was 9.32,relative molecular weight was 26.145 kDa and the molecular formula was C 1218 H 1893 N 301 O 317 S 10.It is much interesting that a signal residing in ER(endoplasmic reticulum)EKKT motif was found in AsSIP2-1.There was 6 trans-membrane domains and no signal peptide in the protein,suggesting it was trans-membrane proteins and belonged to non-secretory protein.The transcriptomic differential expression analysis found that AsSIP 2-1 was significantly down-regulated under drought stress,suggesting that it participated in the response of water shortage.
