摘要
目的采用超高效液相色谱-四极杆-飞行时间质谱(UPLC-Q-TOF-MS)技术对中药复方消痞和胃胶囊中的化学成分进行研究。方法采用Waters CORTECS UPLC C18色谱柱(2.1 mm×100 mm,1.6μm),以乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,柱温35℃,流速为0.3 ml/min,进样量2μl;采用ESI源在正、负离子两种模式下分别进行检测鉴定。通过自建数据库与UNIFI自带数据库结合检索得到初筛化合物,人工复核各化合物的相对保留时间、精确质荷比、特征碎片等信息,并结合相关文献以及化合物裂解规律,鉴定该胶囊的主要化学成分。结果利用UPLC-Q-TOF-MS技术,从该胶囊分离鉴定了46个化学成分,包括皂苷类化合物8个、黄酮类化合物9个、三萜类化合物8个、有机酸类化合物13个等。结论首次采用UPLCQ-TOF/MS联用技术研究消痞和胃胶囊中化学成分,该结果为其质量控制以及进一步的药效物质基础研究奠定了基础。
Objective To qualitatively analyze the chemical constituents of XiaoPiHeWei capsules by UPLC-Q-TOF-MS.Methods The analysis was performed on Waters CORTECS UPLC C18(2.1 mm×100 mm,1.6μm)column.The mobile phase was the acetonitrile-0.1%aqueous formic acid solution in a gradient elution.The flow rate was 0.3 ml/min and the column temperature was 35℃.The mass spectrometric data were acquired in both positive and negative ion modes using ESI.A preliminary search for the identification of compounds was conducted on the self-built database and the UNIFI database.The preliminarily searched constituents were then identified by validating their retention times,accurate mass data,fragment ions and other informations,via the comparison with the data of reference substances and the data in the literature.Results Forty-six compounds were identified,including 8 saponins,9 flavonoids,8 triterpenes and 13 organic acids,etc.Conclusion The results lay the foundation for quality control and further studies on pharmacodynamic substomces of XiaoPiHeWei capsules.
