摘要
目的探讨薯蓣皂苷对过敏性哮喘小鼠炎症反应的影响及对ERK/p38MAPK信号通路的调控作用。方法 SPF级BALB/c小鼠40只,随机分为对照组、过敏性哮喘模型组、薯蓣皂苷组与地塞米松组,每组10只。利用卵白蛋白致敏联合雾化激发法建立小鼠过敏性哮喘模型。HE染色观察肺组织炎症细胞浸润情况;ELISA检测小鼠血清中OVA特异性免疫球蛋白和小鼠肺泡灌洗液中炎性因子表达;Western blot检测各组小鼠肺组织MAPK信号通路相关蛋白ERK1/2、p38 MAPK、JNK、p-ERK1/2、p-p38 MAPK及p-JNK蛋白表达。结果薯蓣皂苷降低过敏性小鼠肺组织炎性浸润,降低血清中OVA特异性IgE含量(P<0.05),降低BALF中炎性因子IL-4、IL-5及IL-13,促进IFN-的表达水平(P<0.05),抑制哮喘小鼠肺组织p-ERK1/2,p-p38 MAPK及p-JNK蛋白的表达水平(P<0.05)。结论薯蓣皂苷抑制过敏性哮喘小鼠炎症反应,与MAPK信号通路相关。
Objective To investigate the effect of diosgenin on inflammatory response and the regulation of ERK/p38 MAPK signaling pathway in the asthmatic mice. Methods Forty SPF grade BALB/c mice were randomly divided into control group, allergic asthma model group(by ovalbumin sensitization), dioscoside group and dexamethasone group, 10 mice in each group. HE staining was used to analyze the infiltration of inflammatory cells in lung tissue. The expression of inflammatory factors in OBA-specific immunoglobulin and mouse alveolar lavage fluid was detected by ELISA. The expression of ERK1/2, p38 MAPK, JNK, p-ERK1/2, p-p38 MAPK and p-JNK was detected by Western blot. Results Diosgenin reduced the inflammatory infiltration of lung tissue and the content of OVA-specific IgE in serum(P<0.05), and the levels of inflammatory factors IL-4, IL-5 and IL-13 in BALF, and upregulated the expression levels of IFN-in allergic mice(P<0.05), but downregulated the expression levels of p-ERK1/2, p-p38 MAPK and p-JNK protein in lung tissue in asthmatic mice(P<0.05). Conclusion Diosgenin could inhibits the inflammatory response of allergic asthmatic mice, which is related to MAPK signal pathway.
