毛竹屑多糖的提取精制及体外抗氧化研究

摘要 5.0 kg竹屑经乙酸乙酯回流脱脂后,烘干,去除有机溶剂,再以水为溶剂,用微波辅助法对脱脂竹屑进行多糖提取。多糖经活性碳脱色、sevage法(氯仿+正丁醇)脱蛋白纯化精制,得精制多糖PPCP。测定精制多糖PPCP在体外对1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟(•OH)自由基的清除能力;结果表明,PPCP在浓度为31 mg/mL时,对•OH自由基的清除率率为53.36%;在PPCP浓度为23 mg/mL时,对DPPH自由基的清除率为59.2%;由此可见,精制多糖DPPH对羟自由基(•OH)和DPPH自由基在体外有较好的清除能力。 5.0 kg bamboo chips were degreased by ethyl acetate refluxing,then dried to remove organic solvents,and then water was used as solvent to extract polysaccharides from defatted bamboo chips by microwave-assisted method.The purified PPCP was obtained by decolorization with activated carbon and deproteinization by Sevage(chloroform+n-butanol).The scavenging capacity of 1,1-diphenyl2-trinitrophenylhydrazine(DPPH)free radical and hydroxyl(∙OH)free radical in vitro was determined by purified polysaccharide PPCP.The results showed that the scavenging rate of∙OH radical reached 53.36%when PPCP concentration was 31 mg/mL,while the scavenging rate of DPPH radical reached 59.2%when PPCP concentration was 23 mg/mL,which showed that DPPH had a good scavenging ability on hydroxyl radicals(∙OH)and DPPH radicals in vitro.
