摘要
目的通过方法学的考察和研究,建立薄层色谱法来鉴别狗骨药材。由于狗骨中含有较多的蛋白质,这些蛋白质水解后的氨基酸是狗骨药用活性的主要成分,薄层色谱法可以很好地分离氨基酸,使之呈现一定的特征色谱图。通过建立完整系统的薄层色谱方法,以控制药材质量,保证用药安全。方法薄层色谱法用于狗骨药材的鉴别,蛋白质在一定条件下充分水解成多种氨基酸,根据不同氨基酸的极性大小,用合适的展开剂在薄层色谱板上进行分离,并喷以显色剂加热使斑点清晰明显,易于观察。结果取狗骨粉末,采用70%乙醇为溶剂超声15 min进行提取,作为供试品溶液。同法制备对照药材溶液和对照品溶液。吸取上述溶液,点样于薄层硅胶G板上,采用正丁醇-冰醋酸-水(3∶1∶1)为展开剂进行展开,喷以茚三酮试液,以105℃加热显色。收集到的10批样品按此方法检验,均得到较好的分离效果,斑点圆整清晰。结论薄层色谱法操作简便,斑点圆整清晰,重现性好,可作为狗骨药材鉴别的方法。
Objective To establish a TLC method for the identification of dog bone medicinal materials.Because there are many proteins in dog bone,the amino acids hydrolyzed from these proteins are the main components of the medicinal activity of dog bone.TLC can separate the amino acids well and make them present a certain characteristic chromatogram.Through the establishment of a complete system of thin-layer chromatography,to control the quality of medicinal materials,to ensure the safety of people's medication.Methods TLC was used for the identification of dog bone medicinal materials.The protein was hydrolyzed to many kinds of amino acids under certain conditions.According to the polarity of different amino acids,a suitable developing agent was used to separate them on the TLC plate,and the spots were clear and easy to observe when the color developing agent was sprayed and heated.Results Dog bone powder was extracted by ultrasound in 70%ethanol for 15 minutes.The same method was used to prepare the control drug solution and the control solution.Take the above solution,spot the sample on the silica gel G plate,use n-butanol-glacial acetic acid-water(3∶1∶1)as the developing agent to develop,spray ninhydrin test solution,and heat at 105℃.The 10 batches of samples collected were checked by this method,and the results were well separated.The spots were round and clear.Conclusion TLC is a simple,round and clear method for identification of dog bone.
