苓泽合剂对痛风性肾病大鼠模型ABCG2、GluT9及URAT1蛋白表达影响 被引量:8

Effect of Lingze Mixture on the Exptession of ABCG2,GluT9 and URAT1 Protein in Rat Model of Gouty Nephropathy

摘要 目的考察苓泽合剂对痛风性肾病模型大鼠ATP结合盒转运蛋白G2(ATP-binding cassette protein2,ABCG2)、葡萄糖转运蛋白9(Facilitated glucose transporter 9,GluT9)及尿酸转运体1(Urate Transporter1,URAT1)蛋白表达的影响,探讨其治疗痛风性肾病的机理。方法 SD大鼠随机分为7组,分别为空白组,模型组,苯溴马隆片组(西药阳性对照),痛风定胶囊组(中药阳性对照药),苓泽合剂高、中、低剂量组(66.6、33.3、16.7 g·kg-1)。利用酵母粉联合腺嘌呤制备大鼠痛风性肾病模型,28 d后,测定其24 h尿量及尿蛋白、肾系数、血清尿酸、肌酐、尿素氮、尿激酶、β2-微球蛋白、胱抑素(Cys C)、尿N-乙酰-β-D葡萄糖苷酶(NAG)含量,蛋白质印迹(Western blot)法检测肾病组织中ABCG2、GluT9及URAT1蛋白表达。结果与模型组比较,苓泽合剂各组尿量、尿蛋白、肾系数、尿酸、肌酐、尿素氮、β2-微球蛋白、CysC及NAG减少,尿激酶增加,肾组织病理改变均有一定减轻,ABCG2蛋白的表达明显增加,GluT9及URAT1蛋白的表达明显降低。结论苓泽合剂对痛风性肾病大鼠具有较好肾保护作用,可能与其上调肾组织中ABCG2,下调GluT9及URAT1蛋白表达有关。 Objective To investigate the effects of Lingze Mixture on the expression of ABCG2,GluT9 and URAT1 in rats with gouty nephropathy,and to explore the mechanism of treatment of gouty nephropathy. Methods Male SD rats were randomly divided into 7 groups:normal control group,model group,positive control group(benzbromarone,0.01 g·kg-1),traditional Chinese positive control group(Tongfengding Capsule,0.86 g·kg-1)high-dose,middle-dose and low-dose Lingze Mixture groups(66.6,33.3,16.7 g·kg-1). The GN rat model was induced by adenine and potassium oxonate. 28 d after modeling,the therapeutic effect and mechanism of Lingze Mixture on gouty nephropathy were studied by detecting the levels of serum uric acid,creatinine,urea nitrogen,urokinase,β2-microglobulin,cystatin(Cys C),urinary n-acetyl-β-D glucosidase(NAG). Renal morphological changes were detected under light microscope,the expressions of ABCG2,GluT9 and URAT1 were detected by Western blot methods. Results Compared with the model group,the number of urine volume,urine protein,renal coefficient,uric acid,creatinine and urea nitrogen,β2-microglobulin decreased significantly in the Lingze Mixture groups. Lingze Mixture treatment obviously increased Urokinase. The pathological changes in renal tissues in gouty nephropathy rats were relieved,the protein expression level of ABCG2 was up-regulated,GluT9 and URAT1 was down-regulated in Lingze Mixture groups. Conclusion Lingze Mixture plays a role in the improvement of renal function in gouty nephropathy rats by up-regulating the protein expression levels of ABCG2,down-regulating GluT9 and URAT1.
