摘要
目的:建立HPLC梯度洗脱法同时测定通脉降糖胶囊中毛蕊异黄酮葡萄糖苷、芒柄花苷、芒柄花素、太子参环肽B、绞股蓝皂苷XLIX、绞股蓝皂苷A和绞股蓝皂苷ⅩⅦ含量的含量。方法:采用Agilent ZORBAX Eclipse XDB-C 18色谱柱(250 mm×4.6 mm,5μm),柱温25℃;流动相:乙腈-0.3%甲酸水溶液,梯度洗脱,体积流量1.0 mL·min^-1;检测波长分别为260 nm(检测毛蕊异黄酮葡萄糖苷、芒柄花苷和芒柄花素)和203 nm(检测太子参环肽B、绞股蓝皂苷XLIX、绞股蓝皂苷A和绞股蓝皂苷ⅩⅦ)。结果:毛蕊异黄酮葡萄糖苷、芒柄花苷、芒柄花素、太子参环肽B、绞股蓝皂苷XLIX、绞股蓝皂苷A和绞股蓝皂苷ⅩⅦ分别在3.76~94.00、2.19~54.75、5.12~128.00、0.86~21.50、7.98~199.50、8.59~214.75、11.96~299.00μg·mL-1线性关系良好(r≥0.9992),平均加样回收率分别为99.50%、97.97%、98.87%、96.93%、100.00%、99.03%和98.94%,RSD分别为0.87%、1.55%、1.04%、1.18%、0.73%、1.49%和0.90%。结论:该方法操作便捷、重复性好,可用于通脉降糖胶囊中多指标性成分的质量控制。
Objective:To establish an HPLC gradient elution method for simultaneous determination of calycosin7-O-β-D-glucopyranoside,ononin,formononetin,heterophyllin B,gypenoside XLIX,gypenoside A and gypenoside ⅩⅦ in Tongmai Jiangtang Capsules.Methods:An HPLC gradient elution method was established and the separation was carried on a Agilent ZORBAX Eclipse XDB-C 18(250 mm×4.6 mm,5μm)chromatographic column,the mobile phase consisted of acetonitrile-0.3%formic acid solution in gradient elution.The flow rate was set at 1.0 mL·min-1 and the column temperature was set at 25℃.The detection wavelength were set at 260 nm for calycosin7-O-β-D-glucopyranoside,ononin and formononetin,203 nm for heterophyllin B,gypenoside XLIX,gypenoside A and gypenosideⅩⅦ.Results:The linear ranges of calycosin7-O-β-D-glucopyranoside,ononin,formononetin,heterophyllin B,gypenoside XLIX,gypenoside A and gypenosideⅩⅦwere 3.76-94.00,2.19-54.75,5.12-128.00,0.86-21.50,7.98-199.50,8.59-214.75,11.96-299.00μg·mL^-1(r≥0.9992),respectively.The average recoveries were 99.50%,97.97%,98.87%,96.93%,100.00%,99.03%,98.94%with the RSDs of 0.87%,1.55%,1.04%,1.18%,0.73%,1.49%and 0.90%,respectively.Conclusion:The method is easy to operate and repeatable,can be used for quality control of multiple indicator components in Tongmai Jiangtang Capsules.
