H7N9亚型禽流感病毒的全基因克隆及HA、NA基因序列分析 被引量:2

摘要 从1份鸡病料.上提取的核酸通过RT-PCR的方法扩增出禽流感病毒的8个基因片段,将片段克隆到pMD--19T质粒上,挑选阳性质粒进行测序,然后对血凝素(HA)和神经氨酸酶(NA)基因序列进行分析。结果发现其HA和NA基因均与H7N9亚型禽流感病毒A/chicken/China/FQ2/2019的同源性最高,位于欧亚分支。HA蛋白裂解位点为PEVP-KRKRTAR*GL,有多个连续的碱性氨基酸插入,符合高致病性禽流感病毒的分子特征。HA受体结合位点发生了G186V突变。NA蛋白没有发生E119G、R152K、H274Y和R292K耐药位点突变,NA蛋白茎杆区69~73位氨基酸发生了5个氨基酸缺失。结论:该病料中的禽流感病毒为H7N9亚型,具有高致病性禽流感病毒的分子特征,对神经氨酸酶抑制剂敏感,无耐药性,存在有感染人的风险和毒力增强的可能性。
