摘要
目的探讨微小RNA-22在胃癌患者血清中的异常表达及临床意义。方法选择我院2018年1月—2019年3月的门诊及住院患者,分别为胃腺癌组82例、胃腺瘤组76例和健康对照组70例,采用实时定量PCR(real-time PCR,RT-PCR)法检测血清中miRNA-22的表达水平。研究miRNA-22表达差异和胃腺癌临床病理参数间的相关性。结果胃腺癌组中,血清miRNA-22相对表达量低于胃腺瘤组、健康对照组,差异存在统计学意义(P<0.05)。而胃腺瘤组和健康对照组miRNA-22相对表达量差异不存在统计学意义(P>0.05)。T3、T4期患者血清中miRNA-22低表达率为93.6%(44/47),高于T1、T2期患者的77.1%(27/35),差异具有统计学意义(P<0.05);有淋巴结转移者低表达率为97.7%(42/43),高于无淋巴结转移者的82.1%(32/39),差异具有统计学意义(P<0.05);而与性别、年龄、分化程度无相关性(P>0.05)。结论miRNA-22在胃腺癌患者血清中的表达下调,且与胃腺癌的临床病理特征密切相关。
Objective To investigate the abnormal expression of miRNA-22 in serum of patients with gastric cancer and clinical significance.Methods From January 2018 to March 2019,82 cases of gastric adenocarcinoma,76 cases of gastric adenocarcinoma and 70 cases of healthy control group were selected as outpatients and inpatients.The expression of miRNA-22 in serum was detected by real-time PCR(RT-PCR).The relationship between miRNA-22 expression and clinicopathological parameters of gastric adenocarcinoma was analyzed.Results The relative expression of miRNA-22 in serum of patients with gastric adenocarcinoma was significantly lower than that of patients with gastric adenocarcinoma and healthy control group(P<0.05).There was no significant difference in the relative expression of miRNA-22 between the two groups(P>0.05).The low expression rate of miRNA-22 in T3 and T4 patients was 93.6%(44/47),significantly higher than that in T1 and T2 patients(77.1%)(27/35),the difference was statistically significant(P<0.05);the low expression rate in patients with lymph node metastasis was 97.7%(42/43),significantly higher than that in patients without lymph node metastasis(82.1%)(32/39),the difference was statistically significant(P<0.05);but there was no significant correlation with gender,age and differentiation(P>0.05).Conclusion The expression of miRNA-22 in serum of patients with gastric adenocarcinoma is down-regulated,which is closely related to the clinicopathological characteristics of gastric adenocarcinoma.
