摘要
研究赤霉素(GA3)浸种对甜叶菊种子萌发的影响,试验先通过测定浸种后溶液的电导率,从50、100、200、400、600 mg·L-1GA3中选择最佳浓度,确定浸种时间为24 h,与清水浸种作比较,对浸种后种子萌发情况、可溶性糖、淀粉以及可溶性蛋白含量进行测定。结果表明,200 mg·L-1GA3浸种后溶液电导率为178.9μs·cm-1,显著低于(P<0.05)其他各浓度下的浸种液,为甜叶菊浸种最佳浓度。在此浸种浓度下与清水浸种相比,发芽率和发芽势分别提高17.45%和17.40%,发霉率下降43.04%;可溶性糖的变化表现不一致,清水浸种呈下降趋势,赤霉素浸种出现波动,但含量高于清水浸种;淀粉含量在0 d测定中赤霉素浸种比清水浸种低0.07 mg·g-1,之后6 d内无明显变化;可溶性蛋白在赤霉素浸种后变化不大,而清水浸种后则呈现显著下降趋势(P<0.05)。综上所述,所选浓度为200 mg·L-1GA3能够促进甜叶菊种子萌发。
This study investigated the effects of gibberellin(GA3) soaking on the seed germination of Steva rebaudiana Bertoni. The conductivity was first tested in order to choose the best concentration of GA3 from 50, 100,200,400 and600 mg·L-1. 24 h was determined as the seed soaking time and compared with water soaking group. After seeking the seeds, germination of seeds, soluble sugar, starch and soluble protein were recorded and tested. Results showed that after 200 mg·L-1 GA3 soaking, the conductivity of the solution was 178.9 μs·cm-1 and it was significantly(P<0.05) lower than the other concentrations of soaking solution. So this concentration was the best concentration.Soaking in this concentration was compared with soaking with fresh water, the germination rate and germination energy were increased by 17.45% and 17.40% respectively. And the mildew rate was decreased by 43.04%. Water soaking and GA3 soaking were not consistent in the change of soluble sugar. The former showed a downward trend and the later showed fluctuation trend. However, the content was higher than the water soaking. On the 0 d, the starch content in GA3 soaking was 0.07 mg·g-1 lower than in water soaking and after the determination there were no obviously changes between two treatments. The soluble protein showed stable trend in GA3 soaking at the next 6 d but the water soaking showed a significant(P<0.05) downward trend. The research showed that 200 mg·L-1 GA3 could promote seed germination of Bertoni.
