摘要
目的探讨白藜芦醇对高同型半胱氨酸血症(HHcy)大鼠心肌细胞凋亡的逆转作用及相关分子机制。方法建立HHcy大鼠模型;超声心动图评估大鼠心功能;苏木素-伊红、Masson和TUNEL染色分析心肌细胞凋亡;TUNEL法检测细胞凋亡;Western印迹确定线粒体凋亡相关因子(NOX)4、细胞色素C和半胱氨酸-天冬氨酸蛋白酶(Caspase)-3表达;透射电镜观察心肌细胞线粒体形态;荧光显微镜观察细胞凋亡;Mitotracker和JC-1染色检测心肌细胞线粒体膜电位。结果HHcy大鼠心肌细胞线粒体NOX4表达升高,细胞色素C渗漏增多,Caspase-3表达显著升高,细胞凋亡数量增多。HHcy大鼠心肌细胞线粒体膜电位显著下降,线粒体结构和功能破坏。予以白藜芦醇可明显抑制高同型半胱氨酸对心肌细胞的促凋亡和线粒体损伤作用。结论白藜芦醇可通过抑制高同型半胱氨酸对心肌细胞的线粒体相关凋亡和线粒体损伤作用,起到心肌细胞保护作用。
Objective To investigate the reverse effect of resveratrol on apoptosis of rat cardiomyocytes induced by homocysteine and its related molecular mechanism.Methods A rat model of homocysteine was established.Cardiac function was evaluated by echocardiography.HE,Masson and TUNEL staining were used to analyze myocardial apoptosis,TUNEL method was used to detect apoptosis;Western blot was used to determine the expression of mitochondrial apoptosis-related factors NOX4,cytochrome C and Caspase-3.The morphology of mitochondria in myocardial cells was observed by transmission electron microscopy.Apoptosis was observed by fluorescence microscope.Mitotracker and JC-1 staining were used to detect mitochondrial membrane potential in cardiomyocytes.Results Mitochondrial NOX4,cytochrome C and Caspase-3 expression and the number of apoptosis increased in cardiomyocytes with high homocysteine.In addition,homocysteine significantly decreased mitochondrial membrane potential and destroyed mitochondrial structure and function.Resveratrol could obviously inhibit the pro-apoptotic and mitochondrial damage effects of homocysteine on cardiomyocytes.Conclusions Resveratrol could protect cardiomyocytes by inhibiting the mitochondrial-related apoptosis and mitochondrial damage caused by homocysteine.
