摘要
研究林泽兰内酯B在大鼠体内的代谢转化过程。采用超高效液相色谱-四极杆-飞行时间质谱,结合PeakView、MetabolitePilot代谢物分析鉴定软件,系统分析林泽兰内酯B的质谱裂解规律,以及在大鼠体内的代谢产物。林泽兰内酯B经过水解、氧化、还原、甲基化等Ⅰ相反应及葡萄糖醛酸化、磷酸化、谷胱甘肽结合等Ⅱ相反应,形成相关系列代谢产物共26种。本方法阐明了林泽兰内酯B在大鼠体内的生物转化规律,为中药材野马追临床应用及新药研发等提供科学参考。
To study the metabolic transformation process of eupalinilide B in rats, we used ultra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry system combined with metabolite analysis and identification software such as PeakView and MetabolitePilot to analyze the mass spectrometry cleavage of eupalinilide B and the metabolites in rats.Eupalinilide B was subjected to phase Ⅰ reaction such as hydrolysis,oxidation and reduction,and phase Ⅱ reaction such as glucuronidation and glutathione binding to form a series of 26 related metabolites.The method clarifies the biotransformation law of eupalinilide B in rats,and provides scientific reference for clinical application of Lindley Eupatorium Herb and research of new drug.
