乳酸链球菌素与L-乳酸对嗜水气单胞菌的协同抑杀作用 被引量:1

摘要 为探讨乳酸链球菌素(Nisin)与L-乳酸对嗜水气单胞菌的协同抑杀作用,分别采用比浊法、活菌计数法、膜电动势探针等检测Nisin与L-乳酸对嗜水气单胞菌(ATCC 35654、CICC 10500两株菌)生长、存活、生物被膜形成以及菌体膜完整性的影响。结果表明,Nisin与低浓度(3.5 mmol/L)L-乳酸协同,显著抑制了两株嗜水气单胞菌的生长,使生长延滞期延长,最大比生长速率下降,成熟期最高活菌数量降低,且抑制效果随Nisin浓度增加而增强。Nisin和L-乳酸对指示菌有协同致死作用,Nisin与L-乳酸协同处理12 h,可显著(P<0.05)降低嗜水气单胞菌活菌数,造成菌体细胞死亡。机理初探表明:低浓度L-乳酸可以使菌株外膜脂多糖(Lipopolysaccharide,LPS)释放,Nisin进一步使菌体膜电动势消散。研究结果为利用Nisin与L-乳酸或二者的产生菌控制水产品中的嗜水气单胞菌提供了理论依据。 To investigate the synergistic inhibitory effect and mechanism of nisin and L-lactic acid against Aeromonas hydrophila,turbidity,plate count method,membrane electromotive force probe were used to detect the influence of nisin and L-lactic acid on growth,survival,biofilm formation,and membrane integrity of Aeromonas hydrophila(ATCC 35654,CICC 10500). The results showed that nisin and low concentration(3.5 mmol/L)of L-lactic acid synergistically inhibited the growth of two strains of Aeromonas hydrophila. And the inhibitory effect strongthened with the increasing of nisin concentration. The growth parameters modeled with a Gomptz equation showed that nisin and L-lactic acid synergistically prolonged the growth lag phase of Aeromonas hydrophila,the maximum specific growth rate decreased,and the maximum viable count of bacteria at stationary phase also decreased. Besides,nisin and L-lactic acid had a synergistic lethal effect on the indicator bacteria. Nisin and L-lactic acid treated for 12 h reduced the viable count of Aeromonas hydrophila. Results of the mechanism indicated that low concentration of L-lactic acid released the outer membrane LPS from the bacterial cells,and nisin further dissipated the membrane potential. The results would provide a theoretical basis for the use of nisin and L-lactic acid or their producing strains in control of Aeromonas hydrophila in aquatic products.
