摘要
目的:探讨泥鳅多糖对UVB诱导的人角质形成细胞(HaCaT)光损伤的保护作用。方法:用不同剂量(10 mJ·cm^-2、20 mJ·cm^-2、30 mJ·cm^-2、40 mJ·cm^-2、50 mJ·cm^-2、60 mJ·cm^-2)的UVB照射细胞,选择合适的UVB照射剂量建立HaCaT细胞的光损伤模型。用不同质量浓度(10 mg·L^-1、20 mg·L^-1、40 mg·L^-1、80 mg·L^-1、100 mg·L^-1)的泥鳅多糖作用于正常的HaCaT细胞,筛选泥鳅多糖的给药浓度。经60 mJ·cm^-2剂量UVB照射HaCaT细胞后,给予浓度为10 mg·L^-1的泥鳅多糖处理,用MTT法检测细胞活性,用分光光度法检测细胞内超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(malondialdehyde,MDA)水平。结果:经60 mJ·cm^-2剂量UVB照射后,与对照组比较,模型组HaCaT细胞活性显著下降,SOD活性显著下降,MDA水平显著升高。40 mg·L^-1以上的泥鳅多糖对细胞增殖有显著的抑制作用,选择10 mg·L^-1的泥鳅多糖作为给药浓度,与模型组比较,泥鳅多糖处理能显著提高细胞活性,提高SOD活性,降低MDA水平,显著减轻了细胞的光损伤程度,差异均有统计学意义(P<0.05)。结论:泥鳅多糖能够通过提高细胞内SOD活性,加速清除活性氧,抑制膜脂过氧化,增强细胞的抗氧化能力,对UVB诱导的HaCaT细胞光损伤有显著的保护作用。
Objective:To investigate the protective effect of Niqiu(Misgurnus anguillicaudatus)polysaccharides(MAP)on UVB induced human keratinocyte(HaCaT)photodamage.Methods:The HaCaT cells were irradiated with different doses(10 mJ·cm^-2,20 mJ·cm^-2,30 mJ·cm^-2,40 mJ·cm^-2,50 mJ·cm^-2,60 mJ·cm^-2)of UVB,and the appropriate UVB irradiation dose was selected to establish the photodamage model of HaCaT cells.Different concentrations(10 mg·L^-1,20 mg·L^-1,40 mg·L^-1,80 mg·L^-1,100 mg·L^-1)of MAP were applied to normal HaCaT cells to screen the effective concentration of MAP.HaCaT cells were irradiated with UVB at a dose of 60 mJ·cm^-2,and then treated with 10 mg·L^-1 MAP.MTT method was used to detect cell viability,and spectrophotometry was used to detect SOD activity and MDA content of HaCaT cells.Results:After irradiation with 60 mJ·cm^-2 dose of UVB,compared with the control group,the HaCaT cell viability decreased significantly,SOD activity decreased significantly and MDA content increased significantly.MAP at the concentration of 40 mg·L^-1 or more significantly inhibited cell proliferation.Therefore,MAP at the concentration of 10 mg·L^-1 was used to treat HaCaT cells in the subsequent experiments.Compared with the UVB model group,MAP treatment can significantly increase cell viability,increase SOD activity,reduce MDA content,and significantly reduce the degree of photodamage to cells.Conclusion:MAP can significantly improve the intracellular SOD activity,accelerate the removal of ROS,inhibit membrane lipid peroxidation,and improve the cell′s antioxidant capacity.Therefore,MAP have a significant protective effect on UVB induced HaCaT cells photodamage.
