地黄饮子对脑出血大鼠灶周内源性神经干细胞再生及Notch1蛋白表达的影响 被引量:6

Effects of Dihuang Yinzi on regeneration of endogenous neural stem cells and expression of Notch1 protein in rats with intracerebral hemorrhage

摘要 目的:研究地黄饮子(补肾填精益髓法)对脑出血大鼠灶周内源性神经干细胞再生及Notch1蛋白表达的影响。方法:72只SD大鼠被随机分假手术组、模型组、中药组,每组24只,以立体定向仪自体血注入法制备大鼠脑出血模型,将地黄饮子灌胃于中药组,利用免疫组织化学方法,观察脑出血大鼠灶周内源性神经干细胞再生及Notch1蛋白表达的动态变化。结果:在同一时间段,与模型组比较,中药组的神经功能评分显著改善(P<0.05),可显著提高DCX、NeuN、Notch1的表达(P<0.01)。结论:地黄饮子可诱导脑出血大鼠灶周内源性神经干细胞再生,并改善神经功能,可能与Notch信号通路有关。 Objective:To study the effects of Dihuang Yinzi(invigorating kidney semen to benefit marrow method)on the regeneration of endogenous neural stem cells and the expression of Notch1 protein in rats with intracerebral hemorrhage.Methods:Seventy-two SD rats were randomly divided into 3 groups,including sham operation group,control group,and Chinese materia medica group,24 rats in each group.Dihuang Yinzi was used in Chinese materia medica group.The expression of endogenous neural stem cells(NSCs)and the dynamic changes of Notch1 protein expression in the cerebral hemorrhage rats were observed by immunohistochemistry.Results:At the same time,compared with the model group,the neurological deficits in the Chinese medicine group were significantly improved(P<0.05),and the expressions of DCX,NeuN,and Notch1 were significantly increased(P<0.01).Conclusion:Dihuang Yinzi can induce regeneration of endogenous neural stem cells and improve neurological function in rats with intracerebral hemorrhage,which may be related to the Notch signaling pathway.
