摘要
目的:研究中药熟地黄不同提取分离部位对体外培养的海马神经干细胞(NSCs)增殖活性的影响。方法:采用系统溶剂法以及柱色谱分离将熟地黄分成不同部位,采用CCK-8法测定熟地黄各部位对体外培养的海马NSCs增殖活性的影响,并筛选出最佳作用浓度。结果:熟地黄正丁醇部位(0.8、0.4μg/mL,P<0.01)、乙酸乙酯部位(0.8μg/mL,P<0.01)和剩余水部位(0.8、0.4、0.2μg/mL,P<0.01)对NSCs增殖活性有明显促进作用,且乙酸乙酯部位的二氯甲烷-甲醇20∶1洗脱样品(0.2μg/mL,P<0.01)及正丁醇部位的10%乙醇洗脱样品(0.4μg/mL,P<0.01)对NSCs增殖的促进效果最佳。结论:熟地黄乙酸乙酯部位、正丁醇部位和剩余水部位可能含有促NSCs增殖的潜在活性成分。
Objective:To study the effects of different parts of Radix Rehmanniae Preparata on the proliferation of hippocampal neural stem cells cultured in vitro.Methods:The Radix Rehmanniae Preparata extract was divided into different parts by systematical solvent method and column chromatography.The effect of various parts of Radix Rehmanniae Preparata on the proliferation of hippocampal neural stem cells was determined by CCK-8 method,and the optimal concentration was screened.Results:There were significant promotion effects on the proliferation of neural stem cells in the parts of n-butanol(0.8,0.4μg/mL,P<0.01),ethyl acetate(0.8μg/mL,P<0.01)and residual water(0.8,0.4,0.2μg/mL,P<0.01).The samples obtained from the ethyl acetate portion eluted with dichloromethane-methanol 20∶1(0.2μg/mL,P<0.01)and the samples eluted from the n-butanol site with 10%ethanol(0.4μg/mL,P<0.01),have the best effects on promoting the proliferation of neural stem cells.Conclusion:The ethyl acetate part,n-butanol part and residual water part of Radix Rehmanniae Preparata may contain potential active ingredients that stimulate neural stem cell proliferation.
