五子衍宗丸对帕金森病小鼠行为学及氧化应激的影响 被引量:13

Effects of Wuzi Yanzong Pills on behavior and oxidative stress in Parkinson’s disease mice

摘要 目的:观察五子衍宗丸对帕金森病(PD)小鼠行为学及氧化应激的影响。方法:C57BL/6雄鼠随机分为正常组、模型组和五子衍宗丸组,采用MPTP诱导建立小鼠PD模型(正常组除外),同时灌胃给予五子衍宗丸组药物干预,每天2次,持续14d。采用步态、爬杆及胶带移除实验评价小鼠运动功能。微量酶标法检测脑组织谷胱甘肽(GSH)含量;TBA法检测脑组织丙二醛(MDA)水平;Western Blot检测脑组织超氧化物歧化酶(SOD)、过氧化氢酶(CAT)蛋白表达。结果:与正常组比较,模型组步态脚掌接触面积显著增多(P<0.01),转头、爬杆及胶带移除时间均显著延长(P<0.01);脑组织GSH、CAT、SOD表达量均显著降低(P<0.01),MDA含量显著升高(P<0.01);与模型组比较,五子衍宗丸组可显著减少步态脚掌接触面积(P<0.05),缩短转头、爬杆及胶带移除时间(P<0.05,P<0.01),显著增加GSH、CAT、SOD表达(P<0.01),降低MDA含量(P<0.01)。结论:五子衍宗丸可明显改善PD小鼠的运动功能,这可能与抑制氧化应激反应有关。 Objective:To explore the effects of Wuzi Yanzong Pills(WYP)on behavior and oxidative stress in Parkinson’s disease(PD)mice.Methods:C57BL/6 male mice were randomly divided into control group,model group and WYP group.The PD model of mice was established by MPTP(except normal group).At the same time,oral administration of WYP was carried out twice a day for 14 consecutive days.Mice motor function was evaluated by gait,stick climbing and tape removal behavioral experiments.The contents of GSH in the brain were detected by microenzyme labeling and MDA by TBA method.The expression of SOD and CAT in the brain were detected by Western Blot.Results:Compared with control group,the gait foot contact area of model group increased significantly(P<0.01),the time of turning round,climbing pole and removing tape prolonged significantly(P<0.01).The expression of GSH,CAT,SOD in brain decreased significantly(P<0.01),and the content of MDA increased significantly(P<0.01).Compared with the model group,WYP group significantly reduced the gait foot contact area(P<0.05),shortened the time of turning round,pole climbing and tape removal(P<0.05,P<0.01),significantly increased the expression of GSH,CAT,SOD(P<0.01),and decreased the content of MDA(P<0.01).Conclusion:WYP can significantly improve the motor dysfunction of PD mice,which may be related to the inhibition of oxidative stress.
