Trendelenburg体位SD大鼠眼内压、血压、视网膜组织形态变化及机制探讨 被引量:2

Effect and mechanism of Trendelenburg position on intraocular pressure,blood pressure,and retinal tissue morphology in SD rats

摘要 目的观察Trendelenburg体位SD大鼠眼内压、血压、视网膜组织形态的变化,并探讨其机制。方法SD大鼠60只,随机分为实验组和正常组各30只。实验组每天上午8∶00~9∶00将大鼠固定于Trendelenburg体位(头低脚高80°),正常组采取平位。固定体位20 min内每5 min测量眼内压1次、共4次;大鼠固定体位安静5 min后测量尾动脉血压。实验重复1周后处死大鼠,采用HE染色观察大鼠视网膜组织形态变化,检测大鼠血清中的氧化应激指标(MDA含量及GSH-Px、SOD活性)。结果实验1周内实验组每日测得眼内压均高于正常组(P均<0.01);同一实验日内,实验组固定体位后5、10、15、20 min眼内压均高于正常组(P均<0.01),实验组固定体位10 min后眼内压达到最大值。实验组收缩压与舒张压均高于正常组(P均<0.01)。实验组视网膜明显增厚、水肿,细胞胞质空亮,层次欠清晰,内层视网膜细胞间隙增大、变厚。实验组血清MDA含量高于对照组,SOD、GSH-Px活性低于对照组(P均<0.01)。结论Trendelenburg体位可升高SD大鼠眼内压和血压,导致视网膜损伤,机制可能与促进氧化应激有关。 Objective To observe the changes of intraocular pressure(IOP),blood pressure and retinal tissue morphology in SD rats in the Trendelenburg position.Methods Sixty SD rats were randomly divided into the experimental group and the normal group(n=30).At the beginning of the experiment,the rats were fixed in Trendelenburg position at 8:00-9:00 am every day,and the rats in the normal group were fixed in the flat position.IOP was measured once every 5 min within 20 min in the fixed position,4 times in total.The caudal artery blood pressure was measured after the rats were stationary for 5 min.The experiment was repeated for a week,and then the rats were sacrificed.The morphological changes of retinal tissues were observed by HE staining,and the indexes of oxidative stress(MDA content,GSH-PX and SOD activity)in the serum of the rats were detected.Results IOP of the experimental group was higher than that of the normal group everyday within a week(P<0.01).Within the same experimental day,IOP at 5,10,15,and 20 min after fixing the position was higher in the experimental group than in the normal group(all P<0.01),and reached the maximum value at 10 min after fixation.Systolic and diastolic blood pressures in the experimental group were higher than those in the normal group(both P<0.01).In the experimental group,the retina was obviously thickened with edema,the cell cytoplasm was empty,the layer was not clear,and the inner retinal cell gap increased and thickened.The content of MDA in the experimental group was higher than that in the control group,while the activities of SOD and GSH-PX in the experimental group were lower than those in the control group(both P<0.01).Conclusions Trendelenburg position could increase IOP and blood pressure of SD rats,cause retinal damage by promoting the oxidative stress in the body.
